Wnt信号传导通路在阿尔茨海默病患者外周血淋巴细胞中的表达及其临床意义

摘　　要：目的从蛋白水平的角度研究阿尔茨海默病(Alzheimer'sdisease,AD)患者淋巴细胞中应激活化蛋白激酶Wnt的表达情况,为该疾病的筛查提供一个靶点。方法收集2012年9月—2013年12月该院门诊就诊的阿尔茨海默病患者30例以及正常对照组30例,征得患者及其家属的同意后,利用简易精神状态量表(MMSE)等综合评价患者的认知障碍程度并记录患者的基本信息。所有患者空腹抽取15ml新鲜静脉血,应用WesternBlotting技术,检测其中Wnt蛋白的表达水平。结果与正常对照组相比,AD患者外周血淋巴细胞中Wnt蛋白表达水平显著升高(P<0.05)。结论本研究从蛋白水平出发,证实了AD患者淋巴细胞中Wnt蛋白的存在及表达,可能为筛查该疾病的可干预靶点提供了一个简便易用的细胞模型。Objective To study the protein level from the angle of the study of Alzheimers disease （Alzhei- met, sdisease, AD＇） should be the expression of activated protein kinase Writ in lymphoeytes of patients with, and provide an intervention for screening for the disease. Methods From September 2012 to December 2013 in authors hospital outpatient treatment in patients with Alzheimer＇s disease （30 cases） and normal control group （30 cases） were selected as subjects, under the consent of the patients and family, the mini mental state examination （MMSE） was used to evaluate the degree of cognitive impairment. Fresh venous blood samples all patients veere taken for determination of the expression of Wnt protein by Western Blotting technology. Results Compared with the normal control group, levels significantlY increased expression of Wnt protein in peripheral blood lymphocytes in patients with AD （P〈0.05）. Conclusions On the expression level of Wnt protein in the lymphocytes of patients with AD, it is confirmed that Wnt protein may provide an easy to use cell model for the screening of the disease as interfere with target point.
