电离辐射环境影响实验小鼠精液质量的研究

摘  要:目的观察不同程度电离辐射状态下小鼠精液质量的变化情况,为研究电离辐射状态影响人类生育能力的机理提供动物实验依据。方法将125只健康雄性成年昆明小鼠随机分为5组,每组25只,其中对照组予以22℃恒温饲养,不做任何处理;电离辐射组下设1 mGy、5 mGy、10mGy、20mGy4个组,分别于辐射后的第7、9、11、14d,每组随机抽取5只小鼠,测定精子DNA及精子顶体完整率的变化情况。结果 1.与对照组比较,各电离辐射组实验小鼠的精液质量均明显异常,且辐射剂量越大,精液质量下降越快,差异具有统计学意义(P<0.01);2.随着电离辐射持续时间的延长,各电离辐射组实验小鼠的精液质量呈现不同程度的异常,差异具有统计学意义(P<0.01)。结论电离辐射可能通过损伤小鼠精子DNA及附属性腺的功能,导致精子DNA及精子顶体完整率出现不同程度的异常,从而影响其生育能力。Objective To buiId experimentaI animaI modeIs with different degrees of ionizing radi-ation state,measuring changes in sperm DNA and sperm acrosome integrity,and to expIore dif-ferent IeveIs of ionizing radiation in case of a state change mouse semen quaIity,to study the ts atus of ionizing radiation on the basis of animaI studies to provide mechanisms affect human fertiIity.Methods The 125 aduIt maIe mice were randomIy divided intof ive groups with 25 in each.The controI group was raised in a temperature of 22℃without any treatment;ionizing radi-ation group consists of 1 mGy,5 mGy,10 mGy,20 mGy groups,from which 5 were taken ran-domIy from the 4 groups respectiveIy in the first 7 th,9 th,11 th,14 th day after irradiation to test sperm DNA and sperm acrosome integrity.Results 1.Compared with the controI group, the se-men quaIity of the ionizing radiation group of mice were significantIy abnormaI,and the greater the radiation dose,the faster decIine in semen quaIity,the difference was statisticaIIy significant ( P〈0.01);2.As the duration of ionization radiation became Ionger,semen quaIity of the mice in ionizing radiation groups showed varying degrees of abnormaIity,the difference was statisticaIIy significant ( P〈0.01) .Conclusion Ionizing radiation may damage sperm DNA and maIe accessory gIand function,Ieading to varying degrees of abnormaIity of sperm DNA and sperm acrosome integrity,thus affecting their fertiIity.
