摘  要:目的通过研究鲨鱼软骨制剂(SCP)对人胃癌MGC-803细胞Bcl-2的影响,探讨其对人胃癌MGC-803细胞增殖和凋亡的影响及可能的作用机制。方法以不同浓度SCP加入体外培养的MGC-803细胞中,MTT法检测SCP对细胞生长抑制作用;Hoechst33342/PI荧光染色分析细胞死亡特征;流式细胞仪测定细胞凋亡水平;免疫细胞化学染色法检测Bcl-2蛋白的表达。结果 SCP明显抑制MGC-803细胞生长,且抑制作用呈浓度和时间依赖性(P<0.05),其24、48和72h的IC 50值分别为1.61,1.04和0.54mg/ml。1mg/ml SCP处理细胞24 h,细胞出现核染色质凝集、凋亡小体等典型凋亡特征,细胞凋亡比例增大呈时间依赖性(P<0.05)。SCP诱导细胞凋亡过程中Bcl-2表达明显降低。结论 SCP抑制人胃癌MGC-803细胞的增殖及诱导细胞凋亡,下调Bcl-2蛋白表达。Objective To investigate the expression of Bcl-2 in human gastric cancer cell line MGC-803 treated by shark cartilage preparation(SCP) and the possible mechanism of gastric cancer cell proliferation and apoptosis induced by SCP. Methods The different concentrations of SCP were added into the cultured MGC-803 cells. The cells growth inhibition was measured by MTT method, morphology and number of cells were determined by Hoechest33342/PI staining and flow cytometry. The expression level of Bcl-2 protein was measured by immunocytochemistry. Results SCP inhibited MGC-803 cells growth in a dose-and time-dependent manner(P〈0.05). The IC value of SCP for MGC-803 cells 50 for 24,48,72 h was 1.61,1.04 和0.54mg/ml,respectively. MGC-803 cells treated with 1mg/ml SCP for 24 h appeared some typical apoptosis characteristics such as nuclei chromatin condensation and apoptotic body, the proportion of apoptosis cells strikingly increased in time dependent manner(P〈0.05). The expression level of Bcl-2 protein in MGC-803 cells treated with SCP was obviously decreased. Conclusion SCP could inhibit the proliferation and induce apoptosis of MGC-803 cells and downregulate the Bcl-2 protein level.
