摘 要:目的通过生物信息学分析预测hsa-mi R-30b靶基因和功能,为深入研究hsa-mi R-30b的生物学功能和调控机制提供理论指导。方法利用Pub Med检索mi R-30b相关文章,通过mi RBase在线工具分析mi R-30b序列和基因组特征。应用mi RWalk综合数据库对hsa-mi R-30b靶基因进行预测,对靶基因集合应用Cytoscape及其插件Bingo进行功能富集分析,应用DAVID数据库进行靶基因信号转导通路富集分析。mi RBase、Pub Med、mi RWalk数据库通过输入hsa-mi R-30b或mi R-30b名称进行检索,Cytoscape及其插件Bingo和DAVID数据库则输入hsa-mi R-30b靶基因进行分析。结果 mi R-30b序列在多物种间具有一定的保守性。mi RWalk综合数据库预测靶基因交集共125个。hsa-mi R-30b靶基因主要富集于生长发育、细胞迁移、细胞周期、胰腺细胞分化、转录调控、神经系统发育等(P<0.01);KEGG生物通路主要富集于Notch信号通路及年轻起病成人型糖尿病、致心律失常性右心室心肌病、小细胞肺癌和前列腺癌疾病通路(P<0.05)。结论 hsa-mi R-30b预测的靶基因集合富集于多个生物学过程及疾病通路,与肺癌、前列腺癌等多种肿瘤密切相关,为hsa-mi R-30b在肿瘤方向的后续研究奠定基础。Objective To predict hsa-mi R-30 b target genes via bioinformatics analysis, and provide theoretical guidance for further investigation of hsa-mi R-30 b related biological function and regulatory mechanisms. Methods Data of mi R-30 b were searched and collected from Pub Med, the sequence and genome characteristics of mi R-30 b were obtained from mi RBase. Target genes of hsa-mi R-30 b were screened through mi RWalk database and intersection of results were performed by Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis. The searching was achieved by entering hsa-mi R-30 or mi R-30 b in mi RBase, Pub Med and mi RWalk databases and entering targeted genes in Bingo and DAVID database. Results The Mi R-30 b sequences were conserved in many species, the predicted target genes from mi RWalk database were 125. The hsa-mi R-30 b were enriched in developmental process, cell migration, cell cycle, pancreatic cell differentiation,transcriptional regulation and nervous system development(P〈 0.01); The KEGG biological pathway mainly enriched in Notch signaling pathway, young maturity onset diabetes, arrhythmogenic right ventricular cardiomyopathy(ARVC), small cell lung cancer and prostate cancer(P 〈0.05). Conclusion It is demonstrated that the target genes predicted by hsa-mi R-30 b are enriched in multiple biological process, which are related with lung cancer and prostate cancer. The outcome may provide guidance for further tumors study.
