大鼠舌黏膜癌变中转化生长因子-β和叉状头转录因子P3的表达变化及相关性研究

摘  要:目的观察在大鼠舌黏膜癌变过程中转化生长因子-β(transforming growth factor-β,TGF-β)和叉状头转录因子P3(forkhead box P3 transcription factor,FOXP3)的mRNA表达变化情况及两者的相关性,初步探讨两者在大鼠舌黏膜癌变中的作用。方法应用前期实验获取的4-亚基硝氧喹啉(4-nitroquinoline-1-oxid,4NQO)诱发大鼠舌黏膜癌变标本,包括正常黏膜组织(对照组)、异常增生组织(异常增生组)和鳞癌组织(鳞癌组)各8例。采用实时荧光定量聚合酶链反应检测各标本中TGF-β、FOXP3 mRNA表达水平,并对二者表达的相关性进行分析。结果 TGF-β、FOXP3的mRNA相对表达量在对照组中分别为3.0±1.6、1.8±0.8,异常增生组中分别为5.2±1.6、3.6±1.0,鳞癌组中分别为13.6±3.0、6.1±2.5。与对照组相比,TGF-β、POXP3在异常增生组中表达升高,差异有统计学意义(P<0.05);在鳞癌组中两者的表达水平进一步增强。随病理分级增加,TGF-β、POXP3的表达呈现递增趋势。并且,在异常增生和鳞癌组中,TGF-β与FOXP3 mRNA表达呈正相关关系(相关系数r=0.686,P<0.001)。结论 TGF-β、POXP3基因表达异常可能与4NQO诱发的大鼠舌黏膜癌变相关。Objective To investigate the mRNA expression of TGF-β and FOXP3 during oral carcinogenesis. Meth-ods The tissues were obtained from 4-nitroquinoline-1-oxide-induced animal models with tongue carcinogenesis estab-lished in early study. Specimens were divided into three groups: normal ( n=8) , dysplasia ( n=8) , and squamous cell carcinoma ( SCC, n=8) . The transcriptional levels of TGF-βand FOXP3 were examined by real time quantitative PCR. Results The mRNA level of TGF-βwas gradually increased from normal mucosa to dysplasia and SCC ( 3. 0 ± 1. 6, 5. 2 ± 1. 6, and 13. 6 ± 3. 0, respectively) . The transcriptional level of TGF-βin dysplasia group and SCC group was sig-nificantly higher than that in the normal group ( P=0. 043, P〈0. 001, respectively) . The mRNA expression of FOXP3 in normal group was1. 8 ± 0. 8, while 3. 6 ± 1. 0 in dysplasia group, and 6. 1 ± 2. 5 in SCC group. The expression of FOXP3 in the dysplasia and SCC group was also significantly higher than that in the normal group ( P =0. 004, P =0. 005, respectively) . Furthermore, the expression of TGF-β was positively correlated with the level of FOXP3 in the multistage of tongue carcinogenesis ( r=0. 686, P〈0. 001) . Conclusion The upregulation of TGF-βand FOXP3 might be associated with disease progression during 4NQO-induced rat tongue carcinogenesis.
