摘  要:目的探讨骨髓间充质干细胞移植对大鼠脑缺血后Notch1-Dll4信号通路的影响。方法 60只SD大鼠随机分为假手术组,MCAO组,BMSC组,DAPT组。制备大鼠MCAO模型,BMSC组及DAPT组分别于造模成功后24 h经侧脑室注入BMSCs或含DAPT的BMSCs,术后4 d、7 d、14 d取脑,计数脑缺血周边区微血管数量,RT-PCR法检测Notch1、Dll4的表达。结果 BMSC组大鼠脑缺血周边区的微血管数量明显多于MCAO组及DAPT组(P<0.05);BMSC组大鼠Notch1、Dll4表达水平较MCAO组及DAPT组明显增高(P<0.05)。结论骨髓间充质干细胞移植可能通过上调Notch1-Dll4信号通路的表达来促进大鼠脑缺血周边区血管新生。Objective To investigate the effects of BMSCs on the expression of Notch1-Dl4 in the brains of rats with focal cerebral ischemia. MethodSixty male adult Sprague-Dawley rats were randomly divided into 4 groups: Sham group, MCAO group, BMSC group, DAPT group. 24 hours after MCAO, BMSCs were injected into the lateral ventricle of rats in BMSC group, and the same dose of BMSCs contained DAPT was given to the rats in DAPT group. Rats are sacrificed at 4 d, 7 d and 14 d after MCAO. The amount of were counted, and the level of Notch1, Dl4 were detected by RT-PCR. ResultsThe amount of microvessels increased in BMSC group (P〈0.05). Compared with the MCAO group, the Notch1, Dl4 expression in ischemic brain was upregulated (P〈0.05).Conclusion BMSCs can improve the by upregulating the expression of Notch1-Dl4 signaling pathway after MCAO in rats.
