摘 要:目的:比较microRNA-106b(miR-106b)在肾癌及癌旁正常组织之间和肾癌细胞系和正常肾细胞系之间的表达水平,并分析其表达水平与临床病理资料之间的关系。方法:采用RT-qPCR方法检测miR-106b表达量在35对肾癌组织和癌旁正常组织中的差异,并运用秩和检验方法分析肾癌组织中miR-106b表达量与患者临床病理特征之间的相关性。培养肾癌细胞系(786-O和ACHN)和正常肾上皮细胞系(HK-2)并采用RT-qPCR方法检测细胞miR-106b表达量。结果:miR-106b在肾癌组织中的表达量明显高于癌旁正常组织(P<0.01),且肾癌组织中miR-106b表达量与肾癌的组织类型有相关性(P=0.049),未发现与其他临床病理资料如年龄、性别、T分期、Fuhrman分级和AJCC临床分期有明显相关关系(P>0.05)。同时,miR-106b表达量在肾癌细胞系中也明显高于正常肾上皮细胞系(P<0.01)。结论:miR-106b在肾癌组织和细胞系中均高表达,且其在肾癌组织中的表达水平与肾癌组织类型相关,提示miR-106b与肾癌的发生和发展相关。Objective:To compare the expression level of miR-106 bbetween renal cancer and adjacent normal kidney tissues(ANTs)and between cells derived from renal cancer and normal kidney and to analyze the relationship between the expression level of miR-106 band the clinicopathology.Method:Through quantitative real-time polymerase chain reaction(RT-qPCR)we determined the expression of miR-106 bin 35paired renal cancer tissues and ANTs and we analysed the relationship between miR-106 bexpression and clinicopathology by rank sum test.We cultivated the renal cancer cell lines(786-O and ACHN)and human normal kidney cell line HK-2and determined the expression level of miR-106 bin cells by RT-qPCR.Result:MiR-106 bwas up-regulated in renal cell carcinoma(RCC)tissues and cell lines compared with control normal tissues and cell line(P〈0.01).And the miR-106 bexpression in RCC tissues was correlated with histological type(P =0.049),but there was no correlation between miR-106 bexpression and other clinicopathological variables such as age,sex,T stage,Fuhrman grade and AJCC staging(P 〉0.05).Conclusion:MiR-106 bwas up-regulated in both renal cancer tissues and cell lines and miR-106 bexpression level was related to histological type of renal cancer.It is suggested that miR-106 bplay apotential role in the tumorigenesis and development.
