摘 要:目的探讨去白细胞过滤后不同部位取样方法、过滤时血液温度、血液采集后贮存时间对白细胞去除效果的影响。方法使用一次性去白细胞滤器血袋采集抗凝全血,将其分成常温组3 h内过滤、常温组放置5 h后过滤、4℃冷藏5 h后过滤3组。测定过滤前后白细胞去除率、剩余WBC和Hct等变化情况。结果 5 h后室温过滤组与室温3 h内过滤组相比(P>0.05);4℃冷藏5 h后过滤组与室温5h后过滤相比,白细胞去除率显著提高(P<0.05)。结论温度对全血中白细胞的滤除有显著影响,适度的冷藏可显著提高白细胞去除率。在保证制备新鲜冰冻血浆的前提下,适当增加冷藏时间对白细胞滤除是有利的。Objective To explore the leukoreduction filter sampling method at different parts and to investigate the impact of blood temperature at filtration and the storage time of blood after collection on the removal of leukocytes. Methods Disposable leukocyte filter bags were used to collect whole blood, which then were divided into three groups: 3h storage and filtration at room temperature, 5h storage and filtration at room temperature, and 4h refrigeration followed by filtration. The leukocyte removal rate, the remaining white blood cell count, hematocrit and other changes were measured before and after filtration. Results Comparing the groups filtered at room temperature stored for 3 hours and 5 hours, the leukocyte removal rate showed no significant difference (P 〉0. 05). However, the group refrigerated at 4℃ for 5h followed by filtration showed a significant increase in the removal of leukocytes ( P 〈 0. 05 ). Conclusion Blood temperature has a significant influence on the filtered whole blood leukocytes and platelets. Moderate refrigeration can significantly improve the leukocyte removal rate. On the premise of preparing fresh frozen plasma, an increase in refrigeration time is beneficial for leukocyte filtration.
