摘 要:目的探讨食管鳞状细胞癌(ESCC)组织中磷脂酰肌醇蛋白聚糖-3(GPC-3)mRNA和蛋白的表达及意义。方法应用反转录-聚合酶链反应(RT.PCR)和免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测50例ESCC组织、19例癌旁不典型增生组织及50例正常食管黏膜组织中GPC-3mRNA和蛋白的表达。结果在食管正常黏膜组织、癌旁不典型增生组织及癌组织中GPC-3mRNA含量依次降低,分别为0.808±0.068、0.813±0.037、0.875±0.052,组间比较差异有统计学意义(F=43.766,P〈0.05);正常黏膜组织及癌旁不典型增生组织中GPC-3蛋白表达均低于ESCC组织,表达量分别为22.0%(11/50)、42.1%(8/19)、70.O%(35/50),组间比较差异有统计学意义(χ2=23.336,P〈0.05);ESCC组织中GPC-3蛋白表达与癌组织的分化程度、TNM分期、浸润深度及有无淋巴结转移密切相关(P〈0.05)。结论ESCC组织中GPC-3mRNA和蛋白表达均增高,其高表达可能与ESCC发生有关。Objective To explore the expression of glypican -3 (GPC -3) mRNA and protein in esophageal squamous cell carcinoma (ESCC) tisuss. Methods The expression of GPC - 3was detected by reverse transcriptase- polymerase chain reaction (RT-PCR) and streptavid-in- peroxidase(SP) immu- nohistochemical methods in 50 cases of ESCC tissue, 19 cases of mucosa adjacent to cancer, and 50 cases of normal esophageal mucosa. Results The relative content of GPC - 3 mRNA depressed by turns in nor- mal mucosa tissue, mucosa adjacent to cancer and cancer tissue which were 0. 808 ±0. 068, 0. 813 ± 0. 037, 0. 875± 0. 052 respectively. There was a statistical significanae in group comparison ( F = 43.766, P 〈 0. 05 ). The positive rate of GPC - 3 protein expression in normal mucosa and mucosa adjacent to canc- er were lower than that of cancer tissue which were 22.0% (11/50), 42. 1% (8/19), 70. 0% (35/50) re- spectively. There was a statistical significance in group comparison (3(2 =23. 336, P 〈0. 01 ). The expres- sion of GPC - 3 protein was closely correlated with the tumor grade, TNM grade, infiltrative depth and lym- phatic metastasis in ESCC tissue (P 〈 0. 05). Conclusion The expression of GPC - 3 mRNA and protein decreased in ESCC tissue, which suggest the GPC - 3 gene may be related to oncogenesis of ESCC tissue.
