摘  要:目的:探讨盐酸普萘洛尔凝胶治疗婴幼儿血管瘤生长的可能机制。方法:建立婴幼儿血管瘤裸鼠移植瘤模型,第45天将16只裸鼠随机分为实验组和对照组。对照组在移植后给予普通鼠食喂养;实验组在对照组基础上患处局部涂抹盐酸普萘洛尔凝胶,每天2次,持续3周。经过3周治疗后,手术切除血管瘤。采用免疫组织化学法检测病灶组织内血管内皮生长因子(VEGF)和caspase-3表达水平的变化,并采用TUNEL染色检测细胞凋亡情况。结果:与对照组比较,实验组应用盐酸普萘洛尔凝胶后血管瘤病灶内VEGF表达水平明显下降(P<0.05),caspase-3的表达及凋亡指数明显升高(P<0.05)。结论:盐酸普萘洛尔凝胶通过诱导caspase-3的表达,抑制VEGF的作用,减少血管发生,从而对婴幼儿血管瘤裸鼠模型移植瘤的生长起到抑制作用。Objective: To explore the underlying mechanisms for treatment of infantile hemangioma by propranolol hydrochloride gel. Method: Establishment of the model of infantile hemangioma xenografts in nude mice, then randomly di- vided them into two groups on the 45th day. The control group (n= 8)were given normal diet and the experimental group (n=8)additionally were applied with propranolol hydrochloride gel twice a day for 3 weeks. After 3 weeks of treatment, the hemangioma was resected. The expressions of VEGF and caspase-3 in the focal tissue were determined by immunohis- tochemistry. At the same time, the apoptosis of cell were also determined and compared using TUNEL staining technique. Result: Compared with control group, the expressions of VEGF was decreased significantly (P〈0.05), while the expres- sions of caspase-3 and apoptosis index were increased significantly (P〈0. 05). Conclusion: Propranolol hydrochloride gel could induce the expressions of caspase-3 and hindrance the effect of VEGF, thus to control angiogenesis, which thereby effectively inhibit infantile hemangioma xenografts in nude mice.
