胃泌素及胃泌素受体基因在胃癌和癌前病变组织中的表达

摘　　要：目的:观察胃泌素(GAS)及胃泌素受体(CCK-BR)基因在胃癌和癌前病变组织中的表达,探讨GAS及CCK-BR与胃癌发生发展的关系。方法:收集行胃镜检查病人胃组织,根据病理学诊断分为慢性萎缩性胃炎组、肠上皮化生组、非典型性增生组及胃癌组,提取并纯化各组胃组织的总RNA,采用Taqman实时荧光定量PCR技术测定胃组织GAS及CCK-BR的表达,比较各类胃疾病胃组织中GAS和CCK-BR表达水平的差异。结果:GAS及其受体CCK-BR在所有患者胃组织中均有表达,在慢性萎缩性胃炎、肠上皮化生、非典型性增生患者胃组织中GAS和CCK-BR的表达均高于胃癌患者(P<0.05),GAS为胃癌组患者的2.37、1.67及1.9倍,CCK-BR为胃癌组患者的1.62、3.66、1.33倍。结论:GAS和CCK-BR在胃癌组织中的表达低于癌前组织。Objective： To observe gastrin and CCK-BR expression in gastric cancer and precancerous lesions and to explore the relationship between gastrin and CCK-BR and the development of gastric cancer. Methods： The gastric tissues of patients undergoing gastroscopy in the digestive endoscopy center were collected. According to the histopathological diagnosis,patients were divided into 4 groups and total RNA was extracted and purified from gastric tissues,then,gastrin and CCK-BR mRNA levels were determined with Taqman real-time quantitative PCR. Results： Gastrin and its receptor CCK-BR expressed in gastric carcinoma and precancerous lesions. The expression of gastrin in gastritis,intestinal metaplasia,atypical hyperplasia group was higher than that in gastric cancer group,with ratio of2. 37,1. 67,1. 9 times of gastric cancer group,the difference was statistically significant（ P〈 0. 05）.The expression of CCK-BR in gastritis,intestinal metaplasia,atypical hyperplasia group was also higher than that in gastric cancer group,with ratio of 1. 62、3. 66、1. 33 times of gastric cancer group,the difference was statistically significant（ P 〈0. 05）. Conclusions： Gastrin and its receptor CCK-BR expression in gastric cancer tissues is lower than those in the precancerous lesions.
