摘  要:利用Wx基因第1内含子第1位的G/T和第6外显子第1 132位的A/C碱基多态性,分别开发分子标记Wx-I和等位基因特异性分子标记Wx-a-F(Wx-b-F)/Wx-a-R,对76份水稻品种Wx基因多态性进行检测,并分析不同基因型与直链淀粉含量的关系。结果表明,供试品种中在Wx基因的这2个多态性位点上检测到了3种基因型(In1G-Ex6A、In1G-Ex6C和In1TEx6A),分别表现为高、中、低直链淀粉含量;籼稻中这3种基因型均存在,而供试爪哇稻和粳稻中分别只检测到In1G-Ex6C和In1T-Ex6A基因型;供试籼型杂交水稻亲本中,恢复系Wx基因以In1T为主,三系保持系均为In1G,而两系不育系同时含有这2种类型,说明两系不育系的Wx基因变异比三系不育系丰富。Utilizing the nucleotide polymorphism at the 1stbase( G / T) of Intron 1 and the 1132thbase( A / C) of Exon 6 of Wx gene,the molecular markers Wx-1 and Wx-a-F( Wx-b-F) / Wx-a-R were respectively developed for detecting the Wx gene polymorphism in 76 rice cultivars,and the relationship was analyzed between the genotype of Wx gene and the level of amylose content. The results showed that among the tested varieties,three genotypes( In1G-Ex6 A,In1G-Ex6 C,and In1T-Ex6A) were detected on the two polymorphic sites of Wx gene,representing the amylose content at high,medium and low level,respectively. All three genotypes were found in the indica rice cultivars,while only In1G-Ex6 C or In1T-Ex6 A was detected in the tested javanica or japonica rice cultivars,respectively. Among the tested indica hybrid rice parents,In1 T was found dominant in Wx gene of the restorer lines,only In1 G was found in that of maintainer lines,but In1 T and In1 G were both found in that of two-line male sterile lines,revealing that two-line male sterile lines are more diversified than three-line male sterile lines in the variation of Wx gene.
