摘 要:目的研究宫颈癌细胞整体甲基化水平与放疗敏感性的关系。方法用甲基转移酶抑制剂DAC处理宫颈癌He La细胞系,然后给予不同剂量放疗照射,用克隆形成实验评价He La细胞的放疗敏感性。结果 2.5和5μM浓度DAC处理过的He La细胞放疗敏感性高于未处理的He La细胞(P<0.01)。5μM浓度DAC处理过的He La细胞放疗敏感性高于2.5μM浓度DAC处理过的He La细胞(P<0.01)。结论 DAC对宫颈癌He La细胞系的放疗敏感性具有增强作用。Objective To study the correlation between overall methylation level of cervical cancer cells and radiosensitivity. Methods Methyltransferase inhibitor DAC was used to treat cervical cancer HeLa cell line, then different doses of radiotherapy irradiation were given, clone formation assay was used to evaluate radiosensitivity of HeLa cells. Results Radiosensitivities of HeLa cells treated by 2. 5 and 5 p.M DAC were statistically significantly higher than that of HeLa cells without treatment ( P〈0.01 ) . Radiosensitivlty of HeLa cells treated by 5μM DAC was statistically significantly higher than that of HeLa cells treated by 2. 5μM DAC ( P 〈 0. 01 ) . Conclusion DAC can enhance radiosensitivity of cervical cancer HeLa cell line.
