摘 要:目的应用基于核磁共振氢谱(1H nuclear magnetic resonance,1H-NMR)的代谢组学方法研究黄连解毒汤对高果糖诱导的胰岛素抵抗(insulin resistance,IR)模型大鼠胰腺功能代谢组变化的影响。方法选取Wistar大鼠24只,适应一周后随机分3组:对照组、模型组和黄连解毒汤组,每组8只。后两组给予100g/L的果糖水喂饲4周,制备IR模型。同时对照组给予等剂量的纯净水。在接下来的4周,黄连解毒汤组除继续给予100g/L的果糖水同时予以黄连解毒汤其水煎剂3.175g/(kg·d)灌胃,对照组和模型组则给予等量纯净水灌胃。8周末,取大鼠胰腺组织,采集胰腺组织提取液的1H-NMR谱,运用主成分分析法(principal component analysis,PCA)分析。结果与模型组相比,黄连解毒汤组胰腺组织中乳酸、丙氨酸、肌酸、胆碱、甘油磷脂胆碱/磷脂胆碱和肌醇升高,而甜菜碱/氧化三甲胺、牛磺酸和肌苷则降低,均具有统计学意义。结论黄连解毒汤能够调控机体能量代谢、降低肝脏损伤及胰腺毒性,本文从分子水平阐明黄连解毒汤对胰岛素抵抗状态下胰腺组织的代谢调控机制。Objective To study the effect ofHuanglian Jiedu decoction(HJD) on the metabonome ofpancreasin insulin resistance (IR) rats with application of nuclear magnetic resonance 1H spectroscopy(1H-NMR). Methods 24 Wistar rats were divided into three groups, i.e. controls, model group and HJD treated group, 8 rats in each group. Last two groups were given 100g/L fructose water for 4 weeks to establish IR model, normal control group was given the equal dose of pure water at the same time. In the next 4 weeks, rats in HJD group were gavaged with HJD water decoction and were given 100g/L fructose water at the same time, the control group and IR model group were garaged withpure water. At the end of the 8th week, pancreatic tissues were obtained and lH-NMR spectra of each sample was performed and analyzed by PCA method. Results In comparison with model group, the pancreatic extracts levels of lactate, alanine, creatine, choline, GPC/PC and myo-inositol increased, and betaine/TMAO, taurine and inosine decreased in the HJD group. Conclusion HJD can regulate the body's energy metabolism, reduce the injury of liver and the toxicity of pancreas. This study elucidates the metabolic mechanism of HJD on the metabonome of pancreasin insulin resistance (IR) rats in molecular level.
