摘　　要：目的构建颗粒蛋白前体(PGRN)的腺病毒表达载体,观察PGRN的过表达对A431细胞增殖及迁移功能的影响。方法构建PGRN过表达的腺病毒载体,通过细胞增殖实验和划痕实验分别研究PGRN过表达在皮肤鳞癌A431细胞的增殖和侵袭中的作用。结果成功构建获得了PGRN过表达的腺病毒,并成功感染了人皮肤鳞癌细胞A431,在A431中过表达PGRN,并通过CCK-8实验和划痕实验可见过表达PGRN对A431细胞的增殖和迁移有明显的促进作用(P<0.05)。结论 PGRN过表达可以促进皮肤鳞癌A431细胞的增殖和迁移,与皮肤鳞癌的发生、发展密切相关。Objective To construct the progranulin（PGRN） in adenovirus vector and observe the influence of over-ex-pression system of PGRN on growth and metastasis of skin squamous carcinoma A431 cells. Methods The PGRN over-expression system in adenovirus vector was constructed. The cell proliferation assay and scratch test were used to explore the role of PGRN over-expression on cell proliferation and migration. Results The PGRN over-expressing adenovirus and over-expressed the PGRN in A431 cells were successfully obtained. PGRN over-expressing significantly promoted the A431 cell proliferation and migration（P〈0.05）. Conclusion Over-expression of PGRN can significantly promote the proliferation and migration of skin squamous carcinoma A431 cells. It is closely related to development of skin squamous carcinoma.
