应用不同细胞浓度观察裸鼠乳腺癌骨转移的造模成功率

摘　　要：目的观察不同细胞浓度进行裸鼠乳腺癌骨转移造模的成功率。方法30只裸鼠随机分为3组，分别予以浓度为1×106／ml、5×106／ml、1×107／ml的细胞悬液0．1ml进行左心室注射，然后观察其死亡情况，42d进行骨ECT扫描及病理检查，确定其骨转移发生情况。结果骨ECT检查显示骨代谢有异常浓聚，病理检查提示有肿瘤细胞浸润，1×106组无裸鼠死亡，造模成功率为0％，5×106组有1只裸鼠死亡，造模成功率30％，1×107组1只裸鼠死亡，造模成功率70％（P〈0．01）。结论裸鼠经左心室注射能有效建立乳腺癌骨转移模型，采用1×107／ml的细胞浓度是进行裸鼠左心室注射建立乳腺癌骨转移模型较为合适的浓度。Objective To observe the success rate of establishing the nude mice＇ s breast cancer bone metastasis model by using different cells＇ concentrations. Methods Thirty female nude mice were divided equally into 3 groups, then injected with MDA-MB-231 cells suspended in 0.1ml PBS at the concentrations of 1 × 106 （ groupl ） , 5 × 106 （ group2 ） 、 1 × 107个ml （ group3 ） into the left ventricles. Deaths following the injection were recorded, and the mice that survived 42 days after the injection were subjected to the ECT scan and pathological examination to determine bone metastasis. Results ECT scan displayed that bone metabolizing had exceedingly accumulated and pathological examination displayed that there were tumor cells in the bone tissues in some mice. No one mice died and the success rate was 0% in the group1. Only one rice died with 30% success rate in the group2 and in the group3, there was only one mice died, the success rate was 70%, P〈0.01. Conclusions We can establish the nude mice＇ s breast cancer bone metastasis model by injecting the MDA-MB-231 cells into left heart ventricles： And found that the suitable concentration was 1 × 107 个/ml cells suspended in 0. lml PBS.
