摘 要:目的探讨七氟醚预处理对大鼠脊髓缺血再灌注损伤时5脂氧合酶(5-LOX)表达的影响。方法雄性成年SD大鼠45只,体重250~300 g,采用随机数字表法,将大鼠随机分为3组(n=15):假手术组(S组)、脊髓缺血再灌注组(I/R组)和七氟醚预处理组(I组)。采用胸主动脉球囊阻断+体循环低血压法制备大鼠脊髓缺血再灌注模型。I组吸入3.4%七氟醚2 h,24 h后制备模型。于再灌注24 h时采用神经功能缺陷评分(NDS)法评价大鼠神经功能,随后处死取脊髓,分别采用Western blot法和RT-PCR法测定5-LOX、p-ERK1/2和t-ERK1/2蛋白及其m RNA的表达水平。应用ELISA检测血清中肿瘤坏死因子-α(TNF-α)的含量。结果与S组比较,I/R组和I组神经功能缺陷评分升高,I/R组5-LOX和p-ERK1/2蛋白及其m RNA表达上调,TNF-α含量升高,I组5-LOX m RNA和p-ERK1/2蛋白及其m RNA表达上调(P〈0.05);与I/R组比05)。结论七氟醚预处理可能通过下调5-LOX表达,抑制ERK1/2信号通路,从而减轻大鼠脊髓缺血再灌注损伤。【Objective】 To investigate the effect of sevoflurane preconditioning on the expression of 5-lipoxygenase(5-LOX) during spinal cord ischemia-reperfusion(I/R) in rats. 【Methods】 Forty-five male adult Sprague-Dawley rats weighing 250~300g were randomly divided into 3 groups(n = 15 each): sham operation group(group S); spinal cord I/R group(group I/R); sevoflurane preconditioning group(group I). The animals were anesthetized with intraperitoneal 4% chloral hydrate 0.1 mg/kg. Spinal cord ischemia was induced by 10 min occlusion of the thoracic aorta combined with controlled hypotension(MAP = 40 mm Hg). The rats inhaled 3.4%sevoflurane for 2 h and spinal cord I/R was produced 24 h later in group I. The neurological deficits were scored at24 h of reperfusion. The animals were then sacrificed. The expression of 5-LOX, p-ERK1/2 and t-ERK1/2 protein and m RNA was detected using Western blot and RT-PCR respectively. TNF-α levels in serum were detected by ELISA. 【Results】 Compared with group S, the neurological deficit score and TNF-α levels in serum were significantly increased in groups I/R and I, the expression of 5-LOX and p-ERK1/2 protein and m RNA was up-regulated in group I/R, and the expression of 5-LOX m RNA and p-ERK1/2 protein and m RNA was up-regulated in group I(P〈0.05). Compared with group I/R, the neurological deficit score was significantly lower, TNF-α levels in serum was lower, and the expression of 5-LOX and p-ERK1/2 protein and m RNA was lower in group I(P〈0.05).【Conclusion】Sevoflurane preconditioning can reduce spinal cord I/R injury by down-regulating the expression of5-LOX and inhibiting ERK1/2 signaling pathway in rats.
