摘  要:通过LPS刺激鸡成纤维细胞系(DF-1),分别在0、2、4、6、8、12、18和24h等8个时间点采集细胞,提取总RNA,并设计相关引物,应用荧光定量PCR方法检测NLRC5信号通路相关基因(NLRC5、NF-κB、MHC-I、IL-6、IL-18、STAT1、STAT3和TLR4)mRNA表达变化规律。结果显示,LPS刺激后,NF-κB、MHC-I、IL-6、IL-18和STAT3的转录水平均呈现先显著下降(P<0.05)后上升的趋势,NLRC5和TLR4先显著上升后下降的趋势(P<0.05),STAT1的mRNA转录水平无显著性变化(P>0.05)。研究表明LPS刺激能够引起鸡DF-1细胞产生免疫反应,鸡NLRC5对NF-κB的转录起负调控作用。We used LPS to stimulate chicken fibroblast cell lines(DF-1)for 0,2,4,6,8,12,18and24 h,respectively.Total RNA was extracted from cells gathered at different time points,then the transcription difference of association genes in NLRC5 signal pathway(NLRC5,NF-κB,MHC-I,IL-6,IL-18,STAT1,STAT3,TLR4)was determined by quantitative real-time RT-PCR.In comparison with the control group,the transcription of some genes induced by LPS showed conspicuously down regulated and then increased gradually,like a‘V'-type transcription trend.The NLRC5 and TLR4gene expression level induced by LPS was significantly up regulated and increased to their peaks at 4and 6hpost-stimulation,respectively.The results showed that LPS stimulation could cause the immune response of chicken fibroblast cells,transcription of chicken NLRC5 on NF-κB plays a negative regulatory role.
