摘 要:目的本实验通过研究血小板源性生长因子(platelete-derived growth factor,PDGF)A,B,C,D在宫颈癌组织中的表达情况,分析其与宫颈癌临床分期和淋巴管新生的关系。方法采用实时荧光定量聚合酶链反应(real-time polymerase chain reaction,RT-PCR)法及免疫组化分析57例宫颈癌组织和20例正常宫颈组织中PDGF-A,B,C,D的表达情况及其与宫颈癌组织中的微淋巴管密度(lymphatic microvessel density,LMVD)的相关性。结果 PDGF-A,B,D在各期癌症组织中的表达水平均明显高于正常组织(P<0.05),而PDGF-C在正常组织与癌症组织中的表达无差异(P>0.05);PDGF-A在宫颈癌组织中表达随着临床分期的增加呈增高趋势,且具有统计学意义(P<0.05),余3个因子在这三组中的表达量均无统计学意义(P>0.05);正常宫颈组织中LMVD明显低于宫颈癌组织(P<0.05),且宫颈癌淋巴结转移组中的LMVD明显高于无转移组(P<0.05);宫颈癌组织中PDGF-A,B,D的表达与LMVD呈正相关性(P<0.05),但是三个因子在有淋巴结转移组与无转移组中的表达无差异(P>0.05)。结论 PDGF-A,B,D在宫颈癌的发生发展中起着重要的作用,同时PDGFs的表达也促进癌组织中淋巴管的新生,宫颈癌组织中淋巴管新生又是其发生淋巴结转移的早期阶段,因此通过阻断PDGFs的表达可能成为治疗宫颈癌的一个新靶点。Objective To study the platelet-derived growth factor(PDGF)A,B,C,D expression in cervical cancer,and analyze its relationship with the clinical staging of cervical cancer and lymphangiogenesis.Methods PDGF-A,B,C,D expression in57 cervical carcinoma and 20 normal cervical tissues and its correlation with lymphatic microvessel density in cervical cancer tissue were analysed by real-time polymerase chain reaction(RT-PCR)and immunohistochemistry.Results The expression levels of PDGF-A,B,D in various cancer tissues were significantly higher than those in normal tissue(P〈0.05),but not different of PDGF-C between normal tissues and cancer tissues(P〈0.05).The expression of PDGF-A in cervical cancer tissues increased along with the increase of clinical stage(P〈0.05).The other three factors were not significantly different among the three groups(P〈0.05).The lymphatic microvessel density(LMVD)in normal cervical tissues was significantly lower than that in cervical cancer tissue(P〈0.05),and LMVD in the cervical lymph node metastasis group was obviously higher than that without metastasis(P〈0.05).PDGF-A,B,D expression and LMVD in the cervical cancer tissue were positively correlated(P〈0.05).These factors were not significantly different between groups with or without lymph node metastasis.Conclusion PDGF-A,B,D in cervical cancer development plays the important role.The expression of PDGFS promotes lymphangiogenesis carcinoma.Lymphangiogenesis in cervical cancer is the occurrence of early stage of lymph node metastasis,so blocking the expression of PDGFS may be a new target for the treatment of cervical cancer.
