摘  要:为了探究MHC基因的多态性与抗病性之间的相互关联作用。采用GPVYZ感染雏鹅,经Triton X-100分级分离MHC,将Sephadex G-200凝胶柱在Bio Gel P-100凝胶柱色谱工作站上串联,分步聚集纯化鹅肝脏MHC,SDS-PAGE分析鉴定。结果显示,初级分离的MHC主带区为30~90 ku、35 ku、20 ku,纯化的鹅肝脏MHC的特征蛋白带是30 ku,33 ku,40 ku,42 ku低分子量蛋白,与MHC类分子重链/α和轻链/β一致,提示重链与轻链折叠形成多态性MHC类分子,可作为致病研究的候选标记基因。YZ0明显缺失84 ku、61 ku蛋白主带,YZ组明显缺失45 ku蛋白峰p2和42 ku蛋白峰p1特征带,新增加34 ku峰p2和38 ku蛋白峰p1带;提示GPV感染与鹅肝脏MHC基因表达蛋白发生相互作用,45 ku、34 ku MHC是GPV的敏感蛋白,42 ku、38 ku蛋白带也与GPV的感染密切相关,MHC重链基因是GPV的易感基因。In order to understand the correlation between the polymorphism of MHC gene and the disease control,the MHCprotein was analyzed from goslings infected with Goose parvovirus(GPV). MHC was isolated and purified from goslings infect-ed with GPV and control group by Triton X-100 fractionation,chromatographies on Sephadex G-200 and Bio Gel P-100. The re-sults showed that there were 30~90 ku,35 ku and 20 ku main protein bands of the primary separation MHC from goslings infect-ed with GPV by SDS-PAGE and biochemical analysis.The characteristic proteins were the low molecular weight protein of 30 ku,33 ku,40 ku and 42 ku which was the purified MHC from goslings infected with GPV.These were the heavy chain and light chainof the folded polymorphism MHC molecules, which were the best candidate marker gene in disease study. The 84 ku and 61 ku main protein band were missed in the primary separation MHC from goslings infected with GPV,and the 45 ku of protein inpeak 2 and 42 ku of protein in peak 1 obviously disappeared. However,there existed new main protein bands of 34 ku in peak 2and 38 ku of protein in peak 1 in the purified MHC from goslings infected with GPV.These indicated that there existed the interac-tion between GPV infection and the protein expression from MHC gene. The 42 ku,and 38 ku protein of MHC are associated withGPV infection, and MHC gene is the susceptible gene to GPV.
