摘 要:目的:运用磁共振灌注成像技术分别检测VD大鼠病程中1、2、3、4周海马区血流变化,并观察脑通汤的干预作用。方法:采用改良的4血管阻断法(4-VO)进行VD造模,Morris水迷宫检测大鼠学习记忆能力,磁共振灌注技术(PWI)检测大鼠VD病程中海马区血流改变及脑通汤干预作用。结果:(1)与模型组及多奈哌齐组比较,脑通汤组r CBF值均升高,差异有统计学意义(均P<0.01)。(2)与模型组比较,脑通汤组、多奈哌齐组大鼠逃避潜伏期明显短于模型组,穿越平台次数明显增多(P<0.01,P<0.05);与多奈哌齐组比较,脑通汤组穿越平台次数较多,有统计学差异(P<0.05)。结论:脑通汤组可较好的改善大脑海马区血流量的变化,可提高VD大鼠学习记忆能力,且这种作用优于盐酸多奈哌齐。Objective:Magnetic resonance perfusion imaging techniques were used to detect the hippocampal blood flow changes of VD rats and find the effect of Naotongtang treatment in the course of 1,2,3,4 weeks. Methods:The rats were modeled by the improved 4-vessel occlusion(4-VO)method. Learning and memory ability of rats were tested by the Morris water maze. 1,2,3,and 4 weeks after modeling,PWI was used to detect the hippocampal blood flow changes of VD rats respectively and the effect of Naotongtang treatment. Results:(1)Compared with the model group,r CBF of the Naotong tang group increased with the statistical difference,(P〈0.01).Compared with the donepezil group,Naotong tang group's r CBF increased significantly,the difference was statistically significant(P〈0.01).(2)Compared with the model group,Donepezil group and Naotong tang group,escape latency was significantly shorter than the model group,times of crossing the platforms increased significantly(P〈0.01,P〈0.05). Compared with the donepezil group,Naotong tang group run across platform more frequently,showing a significant difference(P〈0.05).Conclusion:The brain hippocampal blood flow,and the rats' ability of learning and memory can be better improved by Naotong tang treatment. Moreover,this effect was better than the hydrochloride donepezil.
