摘  要:目的探讨小干扰RNA(siRNA)沉默IKK/NF-κB信号通路对肝星状细胞株HSC-T6细胞增殖的影响。方法根据GenBank数据库IKKβ的c DNA序列,设计构建合成IKKβsiRNA,采用阳性脂质体转染,设立空白对照组、阴性对照组及实验组,采用MTT法测定IKKβsiRNA转染后24、48、72 h细胞的相对数量及活力,流式细胞仪测定细胞周期的变化。结果 (1)与空白对照组和阴性对照组相比,实验组不同时间HSC-T6细胞增殖均受到明显抑制(P<0.05),且这种抑制效应具有时间依赖性;(2)流式细胞仪检测发现实验组G0/G1期细胞比例增多,S期与G2/M期细胞则显著减少(P<0.05)。结论 siRNA沉默IKK/NF-κB信号通路能够明显抑制HSC-T6细胞的增殖,这种抑制作用是通过改变其细胞周期来实现的,具有潜在的逆转肝纤维化的治疗作用。Objective To investigate the effects of small interfering RNA( siRNA) silencing IKK/NF-κB on the proliferation of hepatic stellate cell line T6( HSC-T6). Methods Chemically synthesized siRNA directed against IKKβwas transfected into HSCs by cationic liposome Lipofectamine TM2000. The cells were divided into blank control group,negative control group and experimental group. The cells in the three groups were cultured for 72 hours. The effect of cell proliferation was studied by MTT at 24 hours,48 hours and 72 hours. Flow cytometry was performed to detect the changes of cell cycle. Results( 1) Compared with the negative control group and blank control group,the proliferation of HSCs in experimental group was inhibited significantly at different time( P〈0. 05).( 2) In the G0/ G1 phase,the cells account increased significantly,while the cells account decreased during S and G2/ M phases( P〈0. 05). Conclusion siRNA silencing IKK / NF-κB can prevent HSC-T6 cell from proliferation significantly in vitro and probably by changing the normal cell cycle of T6 cell. This may be a potential strategy for the treatment of hepatic fibrosis.
