摘 要:目的探索紫菜多糖对刀豆蛋白A(ConA)诱导的急性肝损伤的保护作用及相关机制。方法 30只雄性Balb/c小鼠随机分为5组,对照组灌胃给予生理盐水,ConA模型组尾静脉注入20 mg/kg的ConA;紫菜多糖低、中和高剂量组分别灌胃给予0.5、1及2 g/kg的紫菜多糖,连续给药30 d后,尾静脉注入20 mg/kg的ConA。通过酶联免疫吸附测定、蛋白质印迹试验及免疫组化染色等多种方法评价不同剂量的紫菜多糖对ConA诱导的急性肝损伤的保护作用。结果与对照组相比,ConA模型组的ALT、AST及TNF-α、IFN-γ水平明显升高(P<0.05);与ConA模型组比较,紫菜多糖各剂量组的上述各项指标均显著下降(P<0.05),且呈浓度依赖性。ConA模型组肝脏病理切片出现大片水肿、坏死,紫菜多糖高剂量组坏死程度明显减弱(P<0.05)。与对照组相比,ConA模型组Bcl-2表达水平下降而Bax表达水平上升,紫菜多糖高剂量组上述指标表达情况正好与之相反(P<0.05)。结论紫菜多糖能通过降低肝酶及炎症因子的释放从而保护ConA诱导的急性肝损伤,其保护机制可能与抑制Bcl-2及Bax介导的线粒体内源性凋亡有关。Objective To investigate the protective effect and related mechanism of porphyra polysaccharide on concanavalin A(ConA)-induced acute liver injury. Methods Thirty Balb/c mice were randomly divided into 5 groups: control group, ConA model group, porphyra polysaccharide 0.5, 1 and 2 g/kg groups. The protective effect was evaluated by enzyme-linked immunosorbent assay(ELISA), Western Blot and immunohistochemical staining. Results The expression levels of ALT, AST, TNF-α and IFN-γ in ConA model group were obviously higher than those in control group(P 0.05), and those in the drug treatment group with a dose-dependent manner were obviously lower than in the ConA model group(P 0.05). A large area of edema and necrosis appeared in liver pathological sections of ConA model group while a lower extent in the drug treatment groups(P 0.05). The decreased Bcl-2 and increased Bax in the ConA model group were contrary to those in the porphyra polysaccharide 2 g/kg group(P 0.05). Conclusion Porphyra polysaccharide can protect ConA-induced acute liver injury by reducing liver enzymes and infl ammatory cytokines. Its protection mechanism may be associated with inhibited mitochondrial apoptosis mediated by Bcl-2 and Bax.
