黄连素结合光动力学对胃癌细胞MGC-803凋亡的影响

摘　　要：目的:探讨黄连素结合光动力学对胃癌细胞MGC-803凋亡的影响。方法:MTT法检测黄连素、光动力学疗法(PDT)、黄连素联合PDT处理后MGC-803生长抑制率,并应用联合指数分析两药是否具有协同效应。RT-PCR验证MGC-803经黄连素、PDT、黄连素联合PDT处理后各组survivin、bcl-2、p53、bax基因的表达情况。流式细胞术检测各组细胞的凋亡情况。构建胃癌动物模型,体外模拟黄连素联合PDT治疗胃癌疗效观察。结果:MTT法测各处理组细胞生长情况并计算各组细胞抑制率分别为49.63%,62.29%,84.85%;黄连素与PDT联合后有协同效应,抗肿瘤作用更为显著。RT-PCR、Western blot检测各处理组survivin、bcl-2基因蛋白水平均较对照组明显下调,p53、bax则在处理组明显上调;流式细胞术测得黄连素联合PDT组细胞明显凋亡,各早期组凋亡率分别为5.0%,20.4%,33.5%,40.9%;胃癌小鼠体外模拟黄连素联合PDT治疗,肿瘤体积明显减小。结论:黄连素能协同增强PDT促进MGC-803凋亡的作用、PDT抗肿瘤疗效,黄连素有可能成为新一代PDT增敏剂。AIM： To investigate the effect of berberine combined with photodynamic on apoptosis of gastric cancer cell line MGC-803. METHODS：MTT assay was performed to evaluate the proliferation inhibitory effect of berberine,PDT,berberine and PDT on cultured MGC-803 cells. Analyses using Calcu Syn software were performed to determine whether the combination of berberine and photodynamic on apoptosis of gastric cancer cell line MGC-803. RT-PCR and Western blot were used to detect the expression of survivin,bcl-2,p53 and bax. The apoptosis rate of MGC-803 was determined by flow cytometry（ FCM）. Gastric cancer xenograft mice were treated with berberine and PDT. RESULTS：The proliferation inhibitory rate of experimental groups were 49. 63%,62. 29%,84. 85%,respectively. Berberine with photodynamic was more efficacy compared with that of individual berberine.The gene expression of survivin and bcl-2 in experimental groups were significant down-regulation than those of control group. The gene expression of p53 and bax in experimental groups were significant upregulation than those of control group. MGC-803 apoptosis peak appeared after they were treated with berberine,PDT,berberine and PDT,the percentage of apoptosis cells in the early stage were 5. 0%,20. 4%,33. 5%,40. 9%,respectively. The tumor volume was significant decrease after the mouse models of gastric cancer treated with berberine and PDT. CONCLUSION： Bererine can enhance the effect of PDT inducing cell apoptosis in MGC-803,suggest a potential application of photodynamic therapy for tumor.
