摘 要:目的:研究虫草素对体外培养肺癌细胞株A549恶性生物学行为的调节作用及其与相关miRNA表达的关系。方法:取c57小鼠并喂饲虫草素或生理盐水,处死后取血清;培养肺癌细胞中A549,分为对照组(无血清培养基处理)、虫草素组(虫草素喂饲小鼠的血清处理)、顺铂组(顺铂联合生理盐水喂饲小鼠的血清处理)以及联合处理组(顺铂联合虫草素喂饲小鼠的血清处理),检测细胞的生长情况、迁移情况以及miRNA的含量。结果:(1)虫草素组、顺铂组的OD值、发生迁移的细胞数目均低于对照组(均P<0.01);联合处理组的OD值、发生迁移的细胞数目均低于对照组、虫草素组、顺铂组(均P<0.01);(2)顺铂组和虫草素组的miR-126、miR-145、miR-205、miR-429、miR-499a含量均高于对照组(均P<0.05),联合处理组的miR-126、miR-145、miR-205、miR-429、miR-499a含量均高于对照组、顺铂组和虫草素组(均P<0.05);(3)miR-126、miR-145、miR-205、miR-429、miR-499a的模拟物能够降低细胞的OD值、减少发生迁移的细胞数目;(4)miR-126、miR-145的模拟物能够降低Bcl-2的mRNA含量,miR-205、miR-429、miR-499a的模拟物能够降低MMP9的mRNA含量。结论:虫草素能够抑制肺癌细胞株的增殖和迁移过程,miR-126/miR-145-Bcl2途径和miR-205/miR-429/miR-499a-MMP9途径是虫草素发挥调节作用的分子机制之一。Objective:To study the regulation effects of cordycepinon on malignant behavior of lung cancer cell line cultured in vivo and its relationship with related miRNA expression.Methods:c57mice were fed by cordycepin or saline and serum was collected after death;lung cancer cells A549 were cultured and divided into control group(treated with serum-free medium),cordycepin group(treated with serum of mice fed by cordycepin),cisplatin(treated with cisplatin combined with serum of mice fed by saline)and combined treatment group(treated with cisplatin combined with serum of mice fed by cordycepin).Then cell growth,migration and the miRNA contents were detected.Results:(1)OD value and migrating cellsnumber of cordycepin group,cisplatin group were lower than the control group;OD value and migrating cells number of combined treat-ment group were lower than the control group,cordycepin group,cisplatin group;(2)miR-126,miR-145,miR-205,miR-429,miR-499 alevels of cordycepin group,cisplatin group were higher than the control group.miR-126,miR-145,miR-205,miR-429,miR-499 alevels of combined treatment group were higher than the control group,cordycepin group,cisplatin group.(3)miR-126,miR-145,miR-205,miR-429,miR-499 amimics reduced OD values and migrating cellsnumber.(4)miR-126,miR-145 mimics reduce the mRNA levels of Bcl-2,miR-205,miR-429,miR-499 amimics reduce the mRNA levels of MMP9.Conclusion:Cordyceps can inhibit the proliferation and migration of lung cancer cell lines.miR-126/ miR-145-Bcl2 pathways and miR-205/miR-429/miR-499a-MMP9 way is one of molecular mechanisms which cordyceps regulate malignant behavior of lung cancer cell.
