摘  要:为了探讨不同硒源对黄曲霉毒素B1(AFB1)致鸡肝损伤的作用,选用1日龄健康罗丝308肉仔鸡120羽,随机分为4组:空白对照组(饲喂基础日粮)、AFB1对照组(基础日粮+100μg/kg的AFB1)、亚硒酸钠组(基础日粮+100μg/kg AFB1+0.3 mg/kg Na2Se O3)和硒蛋氨酸组(基础日粮+100μg/kg AFB1+0.3 mg/kg Se Met),试验为期42 d。结果表明:1)AFB1能显著降低肉仔鸡的肝重、血清中总蛋白(TP)、肝组织中谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)和总超氧化物歧化酶(T-SOD)活性,显著升高血清中谷草转氨酶(AST)与谷丙转氨酶(ALT)和肝组织中脂质过氧化产物MDA;2)加硒可以显著缓解以上指标的变化且有机硒作用优于无机硒;3)AFB1能显著增加黄曲霉毒素(AF)代谢的关键酶CYP450 1A5的mRNA和蛋白表达水平,而加硒可以抑制1A5的表达。由以上结果表明,硒特别是有机硒,能有效减轻或基本消除AFB1对肉仔鸡肝脏的损害作用。The aim of this study is to explore the effects of different kinds of selenium on aflatoxin B1( AFB1)- induced hepatotoxicity in vivo. One hundred and twenty one- day- old broilers ROSS 308 chickens were divided into four groups,including control group,AFB1( 100 μg/kg) group,Na2 Se O3+ AFB1( 100 μg / kg) group and Se Met + AFB1( 100 μg / kg) group. The experiment lasted 42 days. The results showed that: 1) Due to the addition of AFB1,liver weight of the broilers,serum total protein( TP),the activities of glutathione( GSH),glutathione peroxidase( GSH- Px) and total superoxide dismutase( T- SOD) in the liver were decreased. On the contrary,aspartate aminotransferase( AST) and alanine aminotransferase( ALT) in the serum and lipid peroxidation product MDA in liver tissue were increased; 2) Selenium could significantly reverse the above indices and Se Met had better effect than Na2 Se O3; 3) AFB1 could significantly increase the mRNA and protein levels of CYP450 1A5,a key enzyme of AF,while selenium could inhibit the expression of 1A5. These results suggested that selenium,especially Se Met,can reduce or substantially eliminate the adverse effects of AFB1 on broiler liver.
