摘  要:从新鲜的人肝癌标本上切除肿瘤组织,移植于裸鼠皮下,建立原代移植瘤并连续传代3次,观察移植瘤的成瘤潜伏期、成瘤率、肿瘤体积、肿瘤的侵袭和转移情况。通过HE染色,与临床标本进行组织病理学比较,利用免疫组化方法比较肿瘤干细胞标志物CD90和乙醛脱氢酶1(ALDH1)在临床和动物模型组织中的表达变化。结果显示动物模型肿瘤组织和临床肿瘤组织结构类似;CD90、ALDH1表达一致。建立人源性肝癌移植动物模型,其组织结构稳定,某些特定蛋白表达一致,可成为肝癌基础研究的重要工具。Fresh specimens excised from the patients with liver cancer were transplanted into nude mice subcutaneously to establish the patient-derived liver cancer xenograft models,which could be passaged 3 times continuously. The tumor latency,take rate,tumor growth,invasion and metastasis of these models were recorded meanwhile.Histopathological structure was compared by HE staining in the clinical samples and the passaged samples. The expression status of cancer stem cell markers CD90 and aldehyde dehydrogenase 1( ALDH1) were checked in these samples using immunohistochemical methods. Similar histopathological structure was observed in those samples,and so did the expression of CD90 and ALDH1. These models are valuable for the basic researches in liver cancer,which had the characters of stable histopathological structure and stable expression of some certain proteins.
