摘  要:目的研究小干扰RNA(siRNA)沉默镁离子转运蛋白1(MagT1)的表达对人乳腺癌细胞系MCF-7增殖和凋亡的影响。方法将MCF-7细胞分为MagT1siRNA实验组(A组)、siRNA阴性对照组(B组)和空白细胞对照组(C组)。采用RT-PCR和Western blot法分别检测MagT1mRNA和蛋白表达,MTT法检测细胞增殖情况,流式细胞术检测细胞凋亡和半胱天冬氨酸蛋白酶9(Caspase-9)表达情况。结果与B组和C组相比,A组MagT1mRNA和蛋白表达降低,Caspase-9表达增加,细胞增殖率降低,细胞凋亡率增加(P<0.05)。结论沉默MagT1表达能抑制乳腺癌细胞增殖,促进细胞凋亡;线粒体信号转导途径参与了细胞凋亡过程。Objective To investigate the effects of magnesium transporter 1(MagT1)silenced by small interference RNA(siRNA)on the proliferation and apoptosis of human breast cancer cell line MCF-7.Methods MCF-7cells were divided into three groups of A(transfected with MagT1siRNA),B(transfected with siRNA negative control)and C(blank control).The mRNA and protein expressions of MagT1 were detected by RT-PCR and Western blot,respectively.The cell proliferation was measured by MTT assay.The cell apoptosis and expression of cystein asparate proteinase(Caspase-9)were determined by flow cytometry.Results Compared with groups of B and C,the expressions of MagT1 mRNA and protein were decreased while expression of Caspase-9was increased in group A(P〈0.05).The cell proliferation rate was lower and cell apoptosis rate was higher in group A than those in groups of B and C(P〈0.05).Conclusion Silencing MagT1 expression can inhibit MCF-7cell proliferation and promote its apoptosis,in which mitochondrial signaling pathway participates.
