摘  要:目的研究米诺环素对脂多糖致大鼠认知功能障碍的影响及可能机理。方法雄性Wistar大鼠32只随机均分为对照组(C组)、脂多糖组(L组)、米诺环素25mg/kg组(M1组)和50mg/kg组(M2组)。L组、M1组及M2组大鼠均分别连续7d腹腔注射250μg/kg脂多糖,C组则给予等容积生理盐水。药物干预第7天,M1组和M2组大鼠腹腔分别注射25mg/kg和50mg/kg米诺环素。给药24h后进行Morris水迷宫实验检测大鼠学习记忆能力,随后取海马组织测定环磷酸腺苷应答元件结合蛋白(CREB)及脑源性神经营养因子(BDNF)的含量。结果与C组相比,L组大鼠登台潜伏期延长,海马组织CREB及BDNF下调(P<0.05)。与L组相比,M1及M2组大鼠登台潜伏期缩短,海马组织CREB及BDNF上调(P<0.05)。结论米诺环素可改善脂多糖引起的大鼠认知功能障碍,其作用可能与海马组织中CREB和BDNF的上调有关。Objective To investigate the effects and underlying mechanism of minocycline on lipopolysaccharide-induced cognitive dysfunction in rats.Methods Thirty-two male Wistar rats were equally and randomly divided into 4groups.Rats in groups of L,M1 and M2 were intraperitoneally injected with lipopolysaccharide 250μg/kg per day for 7days and those in group C were given same volume of normal saline instead of lipopolysaccharide.On the 7th day,the rats in groups of M1 and M2were intraperitoneally injected with minocycline 25mg/kg and 50mg/kg,respectively.Twenty-four hours later,the ability of learning and memory of rats was measured by Morris water maze test.The hippocampus was harvested for the determinations of cAMP-response element binding protein(CREB)and brain-derived neurophic factor(BDNF).Results Compared with group C,the escape latency to the plate was prolonged and the expressions of CREB and BDNF in the hippocampus were downregulated in group L(P〈0.05).Compared with group L,the escape latency to the plate was prolonged and the expressions of CREB and BDNF in the hippocampus were upregulated in groups of M1 and M2(P〈0.05).Conclusion Minocycline is able to improve lipopolysaccharide-induced cognitive dysfunction in rats,which is probably related to the upregulation of CREB and BDNF in hippocampus.
