摘 要:目的:通过检测海人酸致痫大鼠海马神经元Caspase 3和Caspase 9蛋白及mRNA表达,探讨姜黄素对癫痫大鼠神经元保护的分子机制。方法:将30只SD大鼠随机分为正常组、海人酸组和姜黄素组;采用海人酸诱导癫痫造模,姜黄素组造模前7天干预给药,采用免疫组化方法检测大鼠海马神经元Caspase 3和Caspase 9蛋白表达,采用RT-PCR方法检测大鼠海马神经元Caspase 3和Caspase 9 mRNA表达。结果:免疫组化结果显示,与正常组海马神经元比较,海人酸组海马神经元数量减少,胞体皱缩,形态不规则,姜黄素干预后的神经元形态均接近正常对照组的神经元数量和形态;海人酸组神经元胞浆中Caspase 3、Caspese 9表达较正常对照组增高,姜黄素组神经元胞浆中Caspase 3、Caspase 9表达较海人酸组降低(P<0.05)。RT-PCR结果显示,海人酸组Caspase 3和Caspase 9 mRNA表达水平与正常组相比均显著升高(P<0.05);姜黄素组与海人酸组相比,Caspase 3和Caspase 9 mRNA表达水平均显著下降(P<0.05)。结论:姜黄素对海人酸致痫大鼠的神经元凋亡具有抑制作用,可能与下调Caspase 3和Caspase9蛋白及mRNA表达有关。Objective: To investigate molecular mechanism for Curcumin to protect neurons through detecting the expressions of Caspase 3 and Caspase 9 proteins and their mRNA of neurons in kainic acid- induced epileptic rats. Methods: 30 SD rats were divided into normal control group,kainic acid group and Curcumin group randomly,models were induced by kainic injection. Immunohistochemical technique was used to detect Caspase3 and Caspase 9 protein expressions of neurons. RT- PCR was used to detect Caspase 3 and Caspase 9 mRNA expressions. Results: The results of immunohistochemistry technique showed that the neurons in kainic acid group showed less numbers,shrinked body and irregular morphology compared with those in the normal group,the neurons in Curcumin group had similar number and morphology to those in normal group. The Caspase 3 and Caspase 9 protein expressions were obviously increased in kainic acid group compared with those in normal group( P〈0. 05),Caspase 3 and Caspase 9 protein expressions were obviously decreased in Curcumin group compared with those in kainic acid group( P〈0. 05). The results of RT- PCR showed that the Caspase 3 and Caspase 9 mRNA expressions increased significantly in kainic acid group than those in normal group( P〈0. 05),while the Caspase 3 and Caspase 9 mRNA expression in Curcumin group decreased significantly to compare with kainic acid group( P〈0. 05). Conclusion: Curcumin can inhibit the apoptosis of neurons in kainic acid- induced epileptic rat by downregulating the Caspase 3 and Caspase 9 proteins and mRNA expressions of neurons.
