IL-37抑制体外培养的SMMC-7721肝癌细胞的增殖、侵袭和迁移

摘　　要：目的探讨白细胞介素37(IL-37)对SMMC-7721肝癌细胞增殖、迁移和侵袭的影响及可能机制。方法 SMMC-7721肝癌细胞分为重组人IL-37(rh IL-37)处理组和对照组,处理组给予(50、100、200、500)ng/m L rh IL-37,对照组给予等体积培养液。CCK-8法检测细胞增殖能力,划痕实验检测细胞的迁移能力,TranswellTM实验检测细胞侵袭能力;Western blot法检测信号转导子与转录激活子3(STAT3)、磷酸化的STAT3(p-STAT3)、基质金属蛋白酶2(MMP-2)的水平。结果 CCK-8实验结果显示rh IL-37可抑制SMMC-7721肝癌细胞的增殖;划痕实验结果证明rh IL-37抑制SMMC-7721肝癌细胞的迁移;TranswellTM实验结果显示,rh IL-37可抑制SMMC-7721细胞的侵袭。Western blot结果表明,rh IL-37处理可下调SMMC-7721细胞p-STAT3、MMP-2的水平。结论 IL-37可抑制肝癌细胞SMMC-7721的增殖、侵袭、迁移,可能与下调p-STAT3、MMP-2表达有关。Objective To analyze the impact of intedeukin-37 （IL-37） on proliferation, migration and invasion of SMMC-7721 hepatoceUular carcinoma cells and the related molecular mechanism. Methods SMMC-7721 cells cultured in vitro were divided into the （50, 100,200,500） ng/mL recombinant human IL-37 （rhlL-37） treated groups and control group. Cell proliferation ability was detected by CCK-8 assay. Wound healing assay was used to observe cell migration ability. TranswellTM assay was performed to test the cell invasion ability. The levels of signal transducers and activators of transcription 3 （STAT3）, phospho-STAT3 （p-STAT3） and matrix metaUoproteinase-2 （MMP-2） were determined using Western blotting. Results CCK-8 assay showed that rhlL-3？ inhibited the proliferation of SMMC-7721 cells. Wound healing assay revealed that rhlL-37 inhibited the migration of SMMC-7721 cells. TranswellTM indicated that rhlL-37 inhibited the invasion of SMMC-7721 cells. Western blotting demonstrated that rhlL-37 down-regulated the expressions of p-STAT3 and MMP-2. Conclusion IL-37 can inhibit the proliferation, migration and invasion of SMMC-7721 cells. It may be related with the down-regulation of p-STAT3 and MMP-2.
