摘 要:目的分析mi R-153在卵巢癌中的表达,并探讨mi R-153的生物学作用。方法荧光定量PCR法检测卵巢癌组织及细胞中mi R-153的表达水平;转染mi R-153类似物(mimics)至SKOV3细胞,MTT法、平板克隆实验及流式细胞仪分别检测mi R-153对细胞增殖及凋亡的影响。结果 mi R-153在卵巢癌组织及细胞中的表达显著下调;转染mi R-153 mimics后细胞生长受到明显抑制,克隆数目减少,细胞早期凋亡率显著增加(P<0.01)。结论 mi R-153在卵巢癌中表达下调;过表达mi R-153可显著抑制SKOV3细胞增殖,促进细胞凋亡。Objective This study aims to analyze the miR-153 expression profile in ovarian cancer, explore the biological functions of miR-153. Methods The relative expression of miR-153 in ovarian cancer tissues and cancer cell lines were detected by real-time quantitative RT-PCR. Introduction of miR-153 mimics to SKOV3 cells. The cell proliferation ability were detected by MTT assay and colony formation, and cell apoptosis was detected by flow cytometry. Results The expression of miR-153 was significantly decreased in ovarian cancer tissues and cell lines; introduction of miR-153 mimics into the SKOV3 cells inhibited cell proliferation, colony formation ability and induced apoptosis (P〈0.01). Conclusion miR-153, which is significantly downregulated in ovarian cancer tissues and cell lines, inhibits cell proliferation and induces apoptosis.
