摘 要:目的观察α-硫辛酸(ALA)对戊四氮(PTZ)点燃大鼠认知功能的保护作用,并探讨其机制。方法 45只健康成年雄性Sprague Dawley大鼠随机分为对照组、癫痫组和治疗组,每组15只。癫痫组和治疗组大鼠给予PTZ隔日腹腔注射建立PTZ点燃模型,治疗组同时给予ALA腹腔注射,利用Morris水迷宫实验检测动物空间学习、记忆能力,采用比色法检测海马组织中丙二醛(MDA)和还原型谷胱甘肽(GSH)水平,2',7'-二氯荧光素二乙酸酯(DCFHDA)探针检测脑组织中活性氧自由基(ROS)水平。结果与对照组比较,癫痫组大鼠水迷宫实验中逃避潜伏期显著延长(P<0.05),在目标象限停留时间显著缩短(P<0.05),海马组织中MDA和ROS水平显著增高(P<0.05),GSH水平显著降低(P<0.05);与癫痫组比较,治疗组大鼠ALA治疗后水迷宫实验中逃避潜伏期显著缩短(P<0.05),目标象限停留时间显著延长(P<0.05),海马组织中ROS和MDA水平显著降低(P<0.05),GSH水平显著升高(P<0.05)。结论氧化应激在癫痫发病中有重要作用,ALA抗氧化治疗能减轻脑组织氧化应激损伤,改善癫痫大鼠的认知功能。Objective To observe the protective effect on cognitive function and the related mechanism of alpha-lipoic acid( ALA) in an epilepsy model by pentylenetetrazole( PTZ)-kindling. Methods The 45 healthy adult male Sprague Dawley rats were randomly assigned into control group,PTZ group and drug therapy group,with 15 rats in each group. PTZ group and drug therapy group were injected intraperitoneally PTZ to establish PTZ-kindling model. ALA was injected intraperitoneally in the drug therapy group. Morris water maze( MWM) was used to evaluate spacial learning and memory function. Spectrophotometry was used to detect the levels of malondialdehyde( MDA) and glutathione( GSH),2’,7’-dichlorodihydrofluorescein diacetate probe was used to detect the level of reactive oxygen species( ROS) in the hippocampi of rats. Results Compared with the control group,the escape latency in the MWM was significantly prolonged and time spent in the target quadrant was shortened significantly in PTZ group( P 〈 0. 05),ROS and MDA levels in hippocampus tissue significantly increased and GSH significanly decreased in PTZ group( P 〈 0. 05). However,compared with the PTZ group,the escape latency in the MWM was significantly shorten and time spent in the target quadrant was prolonged significantly in PTZ group( P 〈 0. 05),ROS and MDA levels in hippocampus tissue significantly decreased and GSH significanly increased in drug therapy group( P 〈 0. 05). Conclusions Oxidative stress plays an important role in the pathogenesis of epilepsy. ALA can effectively protect cognitive function and ameliorate oxidative damage.
