银杏叶提取物联合高压氧对局灶性脑缺血再灌注损伤大鼠外周血清氧化相关物质浓度的影响  被引量:10

摘  要:目的探讨银杏叶提取物联合高压氧对局灶性脑缺血再灌注损伤的保护作用及其机制。方法选取50只清洁级雄性SD大鼠,随机分为假手术组(A组)、模型组(B组)、银杏叶提取物组(C组)、高压氧组(D组)和银杏叶提取物+高压氧组(E组),每组10只。制备大鼠右侧大脑中动脉缺血再灌注模型后,C、E组给予银杏叶提取物,A、B、D组给予相同体积的生理盐水,D、E组在舱内行高压氧治疗约75 min。测定造模2、3、6、24 h后大鼠外周血清甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、活性氧类物质(ROS)、丙二醛(MDA)、血小板活化因子(PAF)浓度的变化。结果 B、C、D、E组大鼠各个时间点外周血清GSH-PX、SOD活性值均低于A组,而ROS、MDA、PAF均高于A组(P<0.05);C、D、E组大鼠各个时间点外周血清GSH-PX、SOD活性值均高于B组,而ROS、MDA、PAF均低于B组(P<0.05);D、E组大鼠各个时间点外周血清GSH-PX、SOD活性值均高于C组,而ROS、MDA、PAF均低于C组(P<0.05);E组大鼠各个时间点外周血清GSH-PX、SOD活性值均高于D组,而ROS、MDA、PAF均低于D组(P<0.05)。除A组外,其余4组GSH-PX、SOD活性均于再灌注后3 h开始下降,6 h达到最低值,24 h开始升高,而ROS、MDA、PAF均于再灌注后3 h开始升高,6 h达到高峰,24 h开始下降(P<0.05)。结论银杏叶提取物联合高压氧通过升高外周血GSH-PX、SOD活性与降低外周血ROS、MDA、PAF来减轻脑缺血再灌注损伤。Objective To investigate the protective effect and mechanisms of extract of Ginkgo biloba leaves combined with hyperbaric oxygen on rats with focal and cerebral ischemia-reperfusion injury. Methods A total of 50 male Sprague Dawley( SD) rats were randomly divided into group A( sham operation group,n = 10),group B( model group,n = 10),group C( extract of Ginkgo biloba leaf group,n = 10),group D( hyperbaric oxygen group,n = 10) and group E( extract of Ginkgo biloba leaf + hyperbaric oxygen group,n = 10). Rat models of right middle cerebral artery of ischemia-reperfusion injury had been established,and then group C and E were treated with extract of Ginkgo biloba leaves,while group A,B and D were treated with same volume of physiological saline,and group D and E were treated with inboard hyperbaric oxygen treatment for 75 min. The concentration changes of glutathione peroxidase( GSH-PX),superoxide dismutase( SOD),reactive oxygen species( ROS),malondialdehyde( MDA) and platelet activating factor( PAF)of peripheral blood at 2ndh,3rdh,6thh and 24 thh after model establishment in all the groups were detected. Results The activity values of serum GSH-PX and SOD of peripheral blood were lower,while the ROS,MDA and PAF values at all time points in group B,C,D and E were higher than those in group A( P 0. 05); the activity values of serum GSHPX and SOD of peripheral blood were higher,while the ROS,MDA and PAF value at all time points in group C,D and E were lower than those in group B( P 0. 05); the activity values of serum GSH-PX and SOD of peripheral blood were higher,while the ROS,MDA and PAF values at all time points in group D and E were lower than those in group C( P 0. 05); the activity values of serum GSH-PX and SOD of peripheral blood were higher,while the ROS,MDA and PAF values at all time points in group E were lower than those in group D( P 0. 05).
