摘  要:目的阐述碳酸酐酶IX(carbonic anhydrase IX,CAIX)在肾细胞癌中的表达及意义。方法挖掘Oncomine数据库中关于CAIX的信息,并对目前数据库中资料进行二次分析,对其在肾癌中的作用进行荟萃分析。结果 Oncomine数据库中共收集了440个不同类型的研究结果,其中关于CAIX表达有统计学差异的研究结果有31个,CAIX表达增高的研究有22个、表达降低的研究有9个。在泌尿系统肿瘤中,膀胱肿瘤中高表达的研究有3个、低表达的有0个,肾癌中高表达的研究有5个、低表达的有0个,前列腺癌中表达没有差异。共有8个研究涉及CAIX在肾透明细胞癌组织和正常组织中的表达,共包括969个样本,与对照组相比,CAIX在肾透明细胞癌中高表达(P=5.79×10-9)。不仅如此,在TCGA RCC的研究中,CAIX的DNA copy number和mRNA表达量与肾透明细胞癌总体生存率存在相关性,即低表达CAIX的患者总体生存率较差,高表达CAIX的患者预后较好(P分别为0.000 7和0.019)。结论我们通过对Oncomine基因芯片数据库中肿瘤相关基因信息的深入挖掘,提出CAIX在肾细胞癌组织中高表达,且与肾癌预后有关,将为肿瘤药物的开发提供重要理论依据。Objective To explore the expression and significance of carbonic anhydrase IX (CAIX) in renal cell carcinoma (RCC) . Methods CAIX data were retrieved from Oncomine RCC database. Results In Oncomine database, there were 440 studies of different types concerning expression of CAIX, of which 31 studies were of statistically significance (22 up-regulated and 9 down regulated). In RCC, 5 were up-regulated and 0 were down-regulated. A total of 8 studies were involved CAIX in RCC tissues and normal tissues, including a total of 969 samples. Overall, CAIX expression in RCC is higher than that in normal tissues (P〈0.05). Moreover, in TCGA study, patients with high expression of CAIX have a better survival (Log-rank P〈0.05). Conclusions CAIX was found highly expressed in RCC, which was also a marker for prostate cancer prognosis.
