摘 要:目的:探讨17β-雌二醇对抗癫痫药物短期应用导致的未成熟脑受体表达降低的保护作用。方法:3 d、7 d、14 d、21 d SD大鼠各7窝,共28窝,随机分为对照组、抗癫痫药物组及雌二醇组,抗癫痫药物组又分为苯巴比妥组、托吡酯组以及联合用药组,每组SD大鼠12只。HE染色了解海马CA1区结构,免疫组化标记海马CA1区谷氨酸受体(GluR1、NR2B),灰度分析检测受体表达情况。结果:各年龄段17β-雌二醇药物保护组较未使用17β-雌二醇保护的抗癫痫药物组GluR1、NR2B受体表达均增多(P<0.01);但各17β-雌二醇保护组受体表达较对照组仍有降低(P<0.01)。结论:17β-雌二醇对抗癫痫药物造成的脑损伤有保护作用,但保护作用尚有限。Objective:To explore the protective effect of 17β-estradiol on the decrease of the receptors in immature brain,which was caused by short- term use of antiepileptic drugs.Methods:There were 7 nests of PD3,PD7,PD14,PD21 SD rats,28 nests in all,which were randomly divided into control group,antiepileptic drug group and estradiol group.Antiepileptic drug group was divided into phenobarbital group,topiramate group and combination group,each group was composed of 12 SD rats.The structure of the neurons was measured by HE staining.Immunohistochemical staining was used to tag the GluR1 and NR2 B receptors,and to detect the expression by gray analysis.Results:The expression of the receptors in estradiol group is higher than antiepileptic drugs group(P〈 0.05),but is lower than in control group(P〈 0.05).Conclusion:17β-estradiol produced a protective effect on immature brain damaged by antiepileptic drugs,providing a new research direction for neural protection to the patients using antiepileptic drugs.
