摘 要:目的:探讨右归饮对磨损颗粒诱导的大鼠假体周围骨溶解的影响。方法:选用40只成年雄性SD大鼠,随机分成4组,每组10只,分别为空白对照组、模型对照组、阿仑膦酸钠组和右归饮组。空白对照组不作任何处理;模型对照组行右侧股骨植入聚乙烯颗粒和钛棒制备聚乙烯颗粒诱导假体周围骨溶解大鼠模型;阿仑膦酸钠组每日空腹阿仑膦酸钠2 mL(蒸馏水与片剂的混悬液)灌胃,剂量为0.083mg/(kg·d),1次/d;右归饮治疗组同模型对照组,并以每100g大鼠灌胃2mL计算给药量,每周3次。8周后取血,然后处死所有实验动物以制备股骨标本,进行各组标本的TRAP染色和血清TRACP5b指标检测。结果:1)血清TRACP5b结果为右归饮和阿仑膦酸钠组可明显降低TRACP5b水平,与模型对照组比较差异有统计学意义(P<0.05)。2)TRAP染色阳性OC的观察和分析显示,光镜下右归饮和阿仑膦酸钠组TRAP阳性OC数均少于模型对照组,差异有统计学意义(P<0.05)。结论:右归饮能够有效抑制聚乙烯颗粒诱导的大鼠假体周围骨溶解。Objective: To study the effect of Youguiyin on periprosthetic osteolysis in rat which induced by polyethylene particles. Methods: All 40 adult male SD rats were randomly divided into blank control group, model control group, alendronate group, and Youguiyin treatment group respectively, 10 animals in each group. No treatment was done in blank control group. In model control group, polyethylene particles and titanium rods were implanted into the right femur of rats. In alendronate treatment group, the rats were treated by intragastric administration of 2 mL alendronate sodium (mixed suspension of distilled water and tablets) once daily, with a dose of 0. 083 mg / (kg . d). In Youguiyin treatment group, the rats were administered 2 mL volume drugs per 100 g through intragastrie administration, three times a week. After 8 weeks of establishing model, blood was obtained and all experimental animals were sacrificed to obtain the femur specimens. Both of TRAP staining and serum TRACP5b were tested. Results: 1) Serum TRACP5h results: the level of TRACP5b in both of Youguiyin treatment group and alendronate treatment group were statistically significantly reduced, compared with the model control group (P〈0.05). 2)TRAP staining of osteoclasts under microscope showed the numberof TRAP-positive osteoclasts in both of Youguiyin treatment group and alendronate treatment group werre significantly decreased compared with the model control group (P〈 0.05 ). Conclusion: Youguiyin can effectively inhibit periprosthetic osteolysis which was induced by polyethylene particles.
