奥沙利铂联合天晴甘美对人胃癌细胞体外增殖的影响

摘　　要：目的探讨奥沙利铂联合天晴甘美对人胃癌细胞SGC-7901体外增殖的影响。方法培养人胃癌SGC-7901细胞株,分为对照组、奥沙利铂组、天晴甘美组及低、中、高剂量天晴甘美联合奥沙利铂组(简称低、中、高剂量联合组)。对照组给予葡萄糖处理,奥沙利铂组和天晴甘美组分别给予不同浓度的奥沙利铂(0、0.5、1.0、2.0、4.0、8.0、16.0μmol/L)和不同浓度的天晴甘美(2、4、8、16、32、64μmol/L)处理,各联合组用4 mg/L奥沙利铂分别联合低、中、高剂量(2、8、32μmol/L)的天晴甘美作用于细胞48h,MTT法检测各组细胞增殖抑制率。结果奥沙利铂组随着奥沙利铂剂量的增高,对肿瘤细胞的抑制作用逐渐增强;天晴甘美组中低剂量(2μmol/L)的天晴甘美对胃癌细胞的增殖无明显抑制作用,较高剂量的天晴甘美表现出抑制胃癌细胞增殖的趋势,但抑制作用不明显;各剂量联合组中随着天晴甘美浓度的增加,对胃癌细胞的抑制作用明显增强。结论天晴甘美能明显增强奥沙利铂对胃癌细胞株SGC-7901的增殖抑制作用,两者具有协同作用,且呈剂量依赖性。Objective To explore the influence of oxaliplatin（OP）combined with magnesium isoglycyrrhizinate（MI）on proliferation of gastric cancer cells in vitro. Methods SGC-7901 cell strain of human gastric cancer was cultivated and divided into six groups as：control group,OP group,MI group,and low-dose,medium-dose and high-dose MI＋OP groups.The cells in the control group were managed with glucose,that in OP and MI groups were treated with different concentrations of OP-0,0.5,1.0,2.0,4.0,8.0,and 16.0μmol/L-and MI-2,4,8,16,32,and 64μmol/L,respectively.That in combined MI＋OP groups were treated with 4mg/L OP and low-dose,medium-dose and high-dose of MI-2,8and 32μmol/L,respectively,for 48 hours,the inhibition rates of the cells in each group were then detected using MTT assay. Results In OP group,the inhibitory action on tumor cells gradually enhanced with the increase of OP dose;in low-dose MI group（2μmol/L）,the inhibitory action was not obvious,in higher-dose groups,there was a trend of enhancement,but not significant.In each combined MI＋OP group,the inhibition notably enhanced with the increase dose of MI plus OP. Conclusion Magnesium isoglycyrrhizinate markedly strengthen the inhibition of oxaliplatin on proliferation of SGC-7901 strain of gastric cancer,both of them show synergetic effect and dose dependent.
