摘 要:目的探讨转录因子E2F1、早期生长反应基因-1(EGR-1)在人肾透明细胞癌(RCCC)组织中的表达及临床意义。方法采用免疫组织化学方法检测48例RCCC、28例癌旁肾组织中E2F1和EGR-1蛋白的表达情况。分析E2F1和EGR-1与患者临床病理特征的关系。RT-PCR方法检测34例RCCC、15例癌旁肾组织中E2F1和EGR-1的mRNA表达情况。结果免疫组织化学方法显示:E2F1在RCCC组织中阳性表达率为明显高于癌旁肾组织(P<0.05)。EGR-1在RCCC组织中的阳性表达率明显高于癌旁肾组织(P<0.05);E2F1与EGR-1蛋白在RCCC中的表达呈正相关(r=0.533,P<0.01),与患者性别、年龄无关(P>0.05),而与RCCC组织学分级及临床分期有关(P<0.05)。RT-PCR方法显示:E2F1在RCCC组织和癌旁肾组织中积分吸光度比值比较,差异有统计学意义(P<0.05)。EGR-1在RCCC组织和癌旁肾组织中积分吸光度比值比较,差异有统计学意义(P<0.05)。结论在RCCC组织中,高表达的E2F1和EGR-1可能与RCCC的发生、发展及浸润转移密切相关。Objective To investigate the expression and clinical significance of E2F1 and EGR-1 in renal clear cell carcinoma (RCCC).Methods Immunohistochemistry was used to detect the expression levels of E2F1 protein and EGR-1 protein in 48 cases of RCCC tissue and 28 cases of cancer adjacent tissues.The correlation between the expressions of E2F1, EGR-1and clinicopathological characteristics of RCCC was analyzed.RT-PCR was used to detect the expression levels of E2F1 and EGR-1mRNA in 34 cases of RCCC tissues and 15 cases of cancer adjacent tissues.Results Immunohistochemistry results showed that the positive expression rate of E2F1 in RCCC tissues was significantly higher than that in cancer adjacent tissues (75%vs 32.1%, P〈0.05).The positive expression rate of EGR-1 in RCCC tissues was significantly higher than that in cancer adjacent tissues (79.2%vs 35.7%, P 〈0.05).The expression of E2F1 was positively correlated to that of EGR-1 protein in RCCC tissues ( r =0.533, P 〈0.01),which was not related with patient’s sex and age ( P 〉0.05),however, which was closely correlated to histological grade and clinical stage of RCCC ( P 〈0.05) .RT-PCR results showed that there was a significant difference in the integral absorbance ratio of E2F1 between RCCC tissues and cancer adjacent tissues (0.2628 ±0.0656 vs 0.1225 ±0.0237, P 〈0.05),moreover, there was a significant difference in the integral absorbance ratio of EGR-1 between RCCC tissues and cancer adjacent tissues (0.2788 ±0.0893 vs 0.1279 ±0.02660, P 〈0.05). Conclusion The high expressions of E2F1 and EGR-1 in RCCC may be closely correlated to the pathogenesis, development, infiltration and metastasis of RCCC.
