摘 要:目的观察2-(2-巯基乙醇)-3-甲基-1,4-萘醌(Compound 5,Cpd5)对人宫颈癌HeLa细胞增殖和凋亡的影响。方法四甲基偶氮唑蓝(MTT)法观察Cpd5对HeLa细胞的生长抑制作用,Annexin V/PI双标记流式细胞术检测Cpd5对HeLa细胞的凋亡诱导,Hoechst-33258染色荧光显微镜观察细胞凋亡形态。结果 10、20、30、40、50μmol/L Cpd5处理HeLa细胞48h,生长抑制率分别为4.23、13.11、35.13、51.37、79.90%。不同时间点(12、24、48和72h)检测,细胞生长抑制率存在剂量-时间依赖关系。流式细胞术检测,30μmol/L Cpd5处理12、24和48h后,细胞凋亡率分别达7.15、16.07、44.16%;50μmol/L组的细胞凋亡率明显高于30μmol/L,且随时间增加而显著增加。结论 Cpd5能以剂量-时间依赖的方式抑制HeLa细胞增殖并诱导调亡,是潜在的抗宫颈癌新药。Objective To investigate the influence of 2-(2-mercaptoethanol)-3-methyl-1,4-naphthoquinone(Compound 5,Cpd5)on human cervical cancer HeLa cells' apoptosis and proliferation.Methods MTT assay was used to test growth inhibition of HeLa by Cpd5.An AnnexinV/PI kit was employed for quantifying apoptotic cells by flow cytometry.Moreover,Hoechst-33258 staining was applied to identify the apoptotic cells.Results Treated by Cpd5 at the concentrations of 10,20,30,40 and 50μmol/L for 48 h,the ratios of the cells growth inhibition were respectively 4.23,13.11,35.13,51.37,79.90%.The proliferation inhibition was investigated after treatment by Cpd5 at different concentrations for 12,24,48 and 72h.The cells were inhibited by Cpd5 in a dose-time-dependent manner.Analyzed by flow cytometry,the apoptotic cells were respectively9.25,20.07 and 56.16% after treatment by 30μmol/L Cpd5 for 12,24 and 48hours.The ratio of apoptotic cells in 50μmol/L group was significantly higher than that in 30μmol/L group.Conclusion Cpd5 inhibits HeLa cells' proliferation and induces apoptosis in a dose-time-dependent manner.Cpd5 is a novel potential anti cervical cancer drug.
