摘  要:目的研究柚皮苷(NRG)能否通过调控核因子-κB(NF-κB)通路保护心肌细胞对抗高糖引起的炎症反应及细胞凋亡。方法应用35 mmol/L葡萄糖(高糖,HG)处理H9c2心肌细胞建立高糖损伤的细胞模型,Western blot法检测NF-κB和cleaved caspase-3的表达水平;ELISA法测定白介素1-(IL-1ββ)、IL-6、和肿瘤坏死因子(TNF-α)的分泌水平;Hoechst 33258核染色荧光显微镜照像检测凋亡细胞。结果高浓度葡萄糖对H9c2心肌细胞具有明显的损伤作用,并能促进心肌细胞内NF-κB的转核(均P<0.01)。预处理均能抑制高糖引起的心肌炎症反应,使IL-1β、IL-6、和TNF-α分泌减少(均P<0.01);能抑制高糖诱导的心肌细胞凋亡,使cleaved caspase-3表达(均P<0.01);还能明显地抑制心肌细胞NF-κB的转核(均P<0.01)。结论 NF-κB通路介导高糖引起的H9c2心肌细胞凋亡和炎症反应,NRG可通过抑制NF-κB通路保护心肌细胞对抗高糖引起的炎症反应和细胞凋亡。Objective To investigate whether naringin(NRG) can protect cardiac cells against high glucose(HG)-induced inflammatory response and cell apoptosis through modulating NF-κB Pathway. Methods H9c2 cardiac cells were treated with 35 mmol/L glucose(high glucose, HG) to establish cardiac cell model with HG injury.Western blot assay was used to measure the expression level of NF-κB and cleaved caspase-3. ELISA kit was used to detect the release levels of interleukin-1β(IL-1β), IL-6 and tumor necrosis factor-α(TNF-α). Cell apoptosis was determined by Hoechst 33258 nuclear staining under a fluorescence microscope. Results HG had significant injury on H9c2 cardiac cells and can promote nuclear translocation of NF-κB in cardiac cells(P〈0.01). Pre-treatment can inhibit HG-induced myocardial inflammation, decrease the secretion of IL-1β, IL-6 and TNF-α(P〈0.01); can inhibit HG-induced cardiac apoptosis, facilitate the expression of cleaved caspase-3(P〈0.01); and can also significantly depress NF-κB nuclear translocation in cardiac cells(P〈0.01). Conclusion NRG protects H9c2 cardiac cells against HG-induced inflammatory response and cell apoptosis by inhibiting NF-κB pathway.
