摘  要:研究离体条件下聚乙二醇(PEG-6000)模拟根际渗透胁迫对马铃薯组培苗生长指标影响,为综合评价马铃薯抗旱能力及筛选抗旱马铃薯种质资源提供参考。以耐旱性不同的3个马铃薯品种脱毒试管苗作为试验材料,研究在0(CK,不含PEG)、2%、4%、6%、8%、10%和12%(W/V)PEG-6000胁迫下3个马铃薯品种株高、茎粗、根长、根数、茎鲜重、根鲜重、茎干重、根干重、茎相对含水量和根相对含水量共10个指标变化,并对这些指标相对值进行主成分和隶属函数分析。结果表明,随PEG-6000胁迫强度增加,不同品种不同处理组培苗生长均受到不同程度抑制作用,各指标值总体上均呈现逐渐降低趋势;在2%PEG-6000胁迫下,费乌瑞它根长显著低于对照,而克新1号和克新18号根长轻微高于对照(CK),但与对照差异不显著;采用主成分分析法提取2个主成分,2个主成分提供的信息量占全部信息量90.982%;通过分析在8%PEG胁迫下各个指标变化以及主成分分析所提取的5个生长指标相对值隶属函数,推测这3个供试马铃薯品种抗旱性由强到弱次序是克新1号、克新18号、费乌瑞它。The growth indices of in vitro plantlets of potato under PEG-6000-induced water stress were studied in order to provide a reference for evaluation of potato drought-tolerance and screening of drought-tolerant germplasm. In vitro plantlets of three potato cultivars were stressed by adding 2%, 4%, 6%,8%, 10% and 12%(W:V) of PEG-6000 to Murashige-Skoog solid medium, respectively, with no PEG-6000 addition as control. Plant length, stem diameter, root length, root number, shoot fresh weight, root fresh weight,shoot dry weight, root dry weight, shoot relative water content, and root relative water content were recorded and analyzed by principal components analysis and subordinate function analysis. With the increase of PEG-6000-induced stress, the growth of in vitro plantlets of potato cultivars was all inhibited to some extent,and showed a decreasing trend. Under 2% PEG-6000-induced stress, the root length of the potato cultivar Favorita was significantly shorter than the control, while the root of Kexin1 and Kexin 18 was slightly longer than the control although no significant difference was detected. With the principal component analysis, the first two principal components were extracted, which account for 90.982% of the total variation. Based on decrease of growth index under 8% PEG-6000-induced stress, and subordinate function analysis of relative values of five indices extracted, the rank of drought-tolerance for these three potato cultivars were concluded as Kexin1Kexin18Favorita.
