摘  要:目的使用巴马小香猪建立颈总动脉支架内再狭窄动物模型,探讨该模型中过氧化物酶增殖物激活受体γ(PPAR-γ)与核转录因子NF-кB和磷脂酰肌醇3激酶/蛋白激酶B(PI3K/Akt)的表达变化。方法采用猪股动脉暴露、颈总动脉球囊扩张及支架释放的方法建立猪颈总动脉支架植入动脉模型,3 m后,数字减影血管造影(DSA)及HE染色分析支架内狭窄情况,免疫组织化学和Western blot印迹检测支架血管段和正常颈总动脉PPAR-γ、PI3K/Akt及NF-кB的表达变化。结果支架植入术后3 m,支架植入组颈总动脉支架内明显狭窄,支架段血管NF-кB和P-Akt表达较正常对照组明显上调,PPAR-γ的表达较正常对照组明显下调。结论本实验使用猪股动脉暴露、颈总动脉球囊过度扩张及支架释放的方法,成功建立猪颈总动脉支架内再狭窄动物模型,支架植入组可能通过上调NF-кB和P-Akt的表达及下调PPAR-γ的表达来导致支架内再狭窄的发生。Objective To use obama small swine to establish animal model of carotid artery stent restenosis,discuss the peroxidase proliferation activated receptor gamma( ppar-gamma,PPAR-γ) with the change of Phosphatidyl inositol 3 kinase / protein kinase B( PI3 K / Akt) and nuclear transcription factor NF-кB. Methods Pig femoral exposure,common carotid artery balloon and stent expansion release method of swine common carotid artery stenting artery model was set up,after 3months,digital subtraction angiography( DSA) and HE staining analyzed the stent stenosis,immunohistochemistry and Western blot detected changed stents carotid artery and normal carotid artery NF-кB、PI3K / Akt and ppar-γexpression.Results Three months after stent implantation experimental carotid artery was significantly narrow,NF-кB and P-Akt expression in experimental carotid artery increased more than that of the normal group,PPAR-γ expressed less than normal group. Conclusion The experiment using swine exposed femoral artery,carotid artery balloon over-expansion and stent release successfully established the animal model of carotid artery stent restenosis. The occurrence of stent may be the increased expression of NF-кB and P-Akt and decreased ppar-gamma expression.
