摘 要:[目的]测定水稻黑条矮缩病毒(Rice black-streaked dwarf virus,RBSDV)安徽分离物全基因组序列,分析RBSDV安徽分离物与RBSDV其他分离物以及斐济病毒属(Fijivirus)其他成员之间的分子差异,为明确RBSDV遗传变异与进化规律提供重要信息.[方法]采用RT-PCR法从安徽玉米粗缩病样本中扩增RBSDV的全基因组,克隆、测序并进行序列分析.[结果]获得RBSDV安徽分离物全基因组序列.序列比对发现,RBSDV安徽分离物与其他RBSDV各个分离物S1到S10的序列相似性较高,为87.6%-99.5%,其中与RBSDV韩国分离物(RBSDV-KOR)序列相似性最高,达94.6%-99.5%;从构建的系统关系树也可以看出,RBSDV安徽分离物和其他所有RBSDV分离物均聚成一个单独的分支.[结论]RBSDV各分离物之间亲缘关系很近.RBSDV安徽分离物与RBSDV-KOR亲缘关系最近,可能起源于相同的祖先.另外,推测RBSDV各个基因组片段可能是协同进化的.[Objectives]The complete genome sequences of Anhui isolate of Rice black-streaked dwarf virus(RBSDV)was determined,and the molecule differences between Anhui isolate and other isolates of RBSDV and other members of Fijivirus were analyzed. The work will provide important information for ascertaining genetic variations and evolutionary laws of RBSDV. [Methods]The complete genomes of RBSDV from Anhui maize sample infected with maize rough dwarf disease were amplified by RT-PCR,and then the complete genomes were cloned,sequenced and analyzed. [Results]The complete genome sequences of Anhui isolate of RBSDV were acquired. Sequence analysis showed that S1 to S10 of Anhui isolate of RBSDV shared higher sequence similarity(87.6%-99.5%)with that of other isolates of RBSDV,and had the highest sequence similarity(94.6%-99.5%)with that of Korean isolate of RBSDV(RBSDV-KOR). Furthermore,from the constructed phylogenetic tree,it could be also found that Anhui isolate of RBSDV and other isolates of RBSDV were clustered into a separate branch. [Conclusions]The relationship among the isolates of RBSDV was very close. Anhui isolate of RBSDV and RBSDV-KOR had the closest relationship,which indicated that they might originate from the same ancestor. Moreover,it could be speculated that the different segments of RBSDV genome were co-evolutionary.
