摘  要:采用反相离子对色谱法建立了一种快速测定猪肉中三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、单磷酸腺苷(AMP)含量的检测方法。采用Waters BEH C18柱作为色谱柱,以20 mmol/L磷酸钾缓冲溶液(含5 mmol/L四丁基溴化铵,pH=6.5)和乙腈作为流动相进行梯度洗脱,采用光二极管阵列检测器进行检测,检测波长为259 nm。结果表明:HPLC可以实现猪肉样品中ATP、ADP、AMP 3种物质的同步检测,平均回收率在80.89%~97.82%,相对标准偏差小于2.81%。We achieved rapid determination of adenosine triphosphate (ATP), adenosine diphosphate (ADP) and adenosine monophosphate (AMP) in pork by using ion-pair reverse phase HPLC method. A Waters BEH C18 column was used as stationary phase. The solution containing 20 mmol/L potassium phosphate buffer and 5 mmol/L tetrabutyl ammonium bromide (pH 6.5) and acetonitrile was applied as mobile phase for gradient elution. Photodiode array detector was employed as a detector, and detection wavelength was fixed at 259 nm. The results showed that ATP, ADP and AMP could be simultaneously detected from pork samples, and the obtained average recoveries ranged from 80.89% to 97.82%. The relative standard deviation was less than 2.81%.
