Fe^2＋对CFA小鼠热激疼痛行为的影响

摘　　要：目的观察鞘内及腹腔注射Fe^2＋对完全弗氏佐剂（complete Freund＇s adjuvant，CFA）小鼠疼痛行为的影响。方法制备CFA小鼠模型。将18只小鼠随机分成3组（n=6）：生理盐水（NS1）组、Fe^2＋50μmol/L（Fe50）组、Fe^2＋250μmol／L（Fe250）组，行鞘内给药；另将18只小鼠随机分为3组（n=6）：NS：组、Fe^2＋1．25mmol／L（Fe1.25）组、Fe^2＋2．5mmol／L（Fe2.5）组，行腹腔给药。观察两种给药方式对小鼠热激痛阈值的影响。结果建模后鞘内给药时，与NS1组相比，Fe50组、Fe250组小鼠的热激痛阈值均显著提高（P〈0．05）；腹腔给药时，与NS2组相比，Fe1.25组、Fe2.5组小鼠的热激痛阈值无明显变化（P〉0．05）。结论鞘内注射Fe^2＋对CFA小鼠具有镇痛效应，腹腔注射Fe^2＋镇痛效果不明显。Objective To investigate the effects of intrathecal and intraperitoneal injections of ferrous ions （ Fe^2 ＋ ） on complete Freund＇s adjuvant （CFA） - induced chronic inflammatory pain in mice. Methods A mouse model of CFA was established. A total of 18 mice were randomly divided into the following three groups （n =6） for intrathecal administration ： a normal saline （ NSl ） group, a 50 μmol/L Fe^2＋ （ Fe50 ） group and a 250 μmol/L of Fe^2＋ （ Fe250 ） group. Meanwhile, another 18 mice were randomly divided into the following three groups （n = 6） for intraperitoneal administration： a normal saline （NS2） group, a 1.25 μmol/L Fe^2＋ （Fe1.25） group and a 2.5 μmol/L of Fe^2＋ （Fe2.5） group. The effects of Fe^2＋ on the threshold of thermal stimulation pain （ represented by the thermal withdrawal latency, TWL） in mice of both groups were observed. Results After intrathecal administration, both the Fe50 and Fe250 groups produced remarkably longer TWL than the NS1 group （P 〈 0.05）. However, after intraperitoneal administration, no significant difference was found as to TWL among the three groups （ P 〉 0. 05 ）. Conclusion Intrathecal injection of Fe^2 ＋ has analgesic effects on CFA mice. In contrast, Fe^2＋ can not produce obviously analgesic effects through intraperitoneal injection.
