摘  要:以30日龄健康陆川仔公猪为研究对象,采用免疫组织化学方法及半定量RT-PCR对中枢神经系统的GnIH进行观察定位,从形态学角度及分子水平为深入研究GnIH的生理功能奠定理论基础。免疫组织化学结果表明,在陆川猪中枢神经系统中,大脑皮质、小脑、海马、嗅球、脑桥、中脑、延髓和脊髓均有GnIH免疫阳性神经元分布,但胞体形态多样,大小不一。半定量RT-PCR结果显示,GnIH mRNA在延髓表达量最高,其次为中脑,嗅球最低,其余为中等表达。GnIH免疫阳性神经元及其纤维在中枢神经系统的分布情况与GnIH mRNA的表达分布一致,进一步印证了GnIH在陆川公猪中枢神经系统的广泛分布。Immunohistochemistry(IHC)and semi-quantitative RT-PCR were used to detect the distribution of GnIH in the central nervous system of 30-day-old male Luchuan piglets,which lay a theoretical basis for further research on the physiological functions of GnIH from the perspective of morphology and molecular level.IHC results showed that GnIH immune positive neurons distribution in central nervous system,for instance cerebral cortex,cerebellum,hippocampus,olfactory bulb,pons,midbrain,medulla oblongata and spinal cord of Luchuan piglets,but the GnIH-ir cell bodies and sizes morphological were diversity.The results of semi-quantitative RT-PCR showed that the expression of GnIH mRNA in the medulla oblongata was the highest among central nervous system organizations,followed by midbrain,the lowest in the olfactory bulb,and the medial in other organizations.The results of IHC and semi-quantitative RT-PCR were coincident,which further confirmed widely distribution of GnIH in the central nervous system of Luchuan piglets.
