摘 要:[目的]探讨西黄丸含药血清与肿瘤坏死因子相关细胞凋亡诱导配体(TNF related apoptosis inducing ligand,TRAIL)单独或联合对人胆管癌细胞RBE增殖和凋亡的影响及其机制。[方法]制备高、中、低剂量西黄丸含药血清;人胆管癌细胞RBE分别用西黄丸含药血清、人重组可溶性TRAIL蛋白(rsTRAIL)及两者联合处理;MTT法检测细胞增殖;流式细胞术检测细胞凋亡;分别用免疫荧光染色结合流式细胞术和RT-PCR检测TRAIL R1/DR4和TRAIL R2/DR5在细胞表面及mRNA水平的表达。[结果]高剂量西黄丸含药血清可以抑制RBE细胞增殖,诱导其凋亡,较对照组差异有统计学意义(P<0.01);中、低剂量西黄丸含药血清未见上述抑瘤作用;rsTRAIL以剂量依赖性方式抑制RBE细胞增殖,诱导其凋亡;低剂量西黄丸含药血清联合rsTRAIL处理后,RBE细胞增殖抑制率和凋亡率明显高于单用rsTRAIL组,比较差异有统计学意义(P<0.05)。低剂量西黄丸含药血清单独或联合rsTRAIL作用于RBE细胞36h后,死亡受体TRAIL R1/DR4和TRAIL R2/DR5在细胞表面和mRNA水平的表达均较对照组细胞明显增强,差异均有统计学意义(P<0.01)。[结论]西黄丸含药血清增强TRAIL对人胆管癌细胞RBE的增殖抑制和凋亡诱导效应,其机制可能与西黄丸含药血清上调RBE细胞TRAIL死亡受体表达相关。[Objective]To investigate the effects of serum containing Xihuang pill,tumor necrosis-related apoptosis inducing ligand(TRAIL),or their combination on the proliferation and apoptosis in human cholangiocarcinoma RBE cells and the mechanisms.[Methods]High,middle and low-dose of serum containing Xihuang pill was made.Human cholangiocarcinoma RBE cells was treated with serum containing Xihuang pill,human recombinant soluble TRAIL(rsTRAIL),or their combinations.The proliferation and apoptosis of RBE cells were analyzed by MTT assay and flow cytometry(AnnexinV-FITC/PI double stainingassay).The expressions of TRAIL R1/DR4 and TRAIL R2/DR5 on the cell surface were determined by immunofluorescence staining and flow cytometry analysis;the mRNA expressions of these molecules were detected by reverse transcription-polymerase chain reaction(RT-PCR).[Results]The serum containing Xihuang pill of high dose significantly inhibited proliferation and induced apoptosis in RBE cells,compared with the control group(P〈0.01),but the serum of middle and low-dose did not.RsTRAIL remarkably inhibited proliferation and induced apoptosis in RBE cells,in a dose-dependent manner.The growth inhibition rate and apoptosis rate of the RBE cells treated with the combination of serum containing Xihuang pill of low dose and rsTRAIL were higher compared with the cells treated with rsTRAIL alone(P〈0.05).After the treatment by serum containing Xihuang pill,or in combination with rsTRAIL,for thirty-six hours,the expressions of TRAIL R1/DR4 and TRAIL R2/DR5 on the cell surface were significantly higher as compared to the control group,so was their mRNA expression level(P〈0.01).[Conclusion]Serum containing Xihuang pill potentiated the growth inhibition and apoptosis induction effects of TRAIL on human cholangiocarcinoma RBE cells and the up-regulation of TRAIL R1/DR4 and TRAIL R2/DR5 expressions on the cell surface may be involved in the synergistic effect.
