穴位埋线对大鼠实验性自身免疫性葡萄膜炎治疗作用的研究

摘　　要：目的探讨穴位埋线对大鼠实验性自身免疫性葡萄膜炎的治疗作用。方法 Liwes大鼠48只,采用随机数字表法分为对照组、模型组、穴位埋线组,每组16只。将光感受器间维生素A类结合蛋白(IRBP)、结核杆菌(TB)和完全弗氏佐剂(CFA)磷酸缓冲盐溶液(PBS)制成乳糜液后,在双足后垫及背部两侧皮下注射。穴位埋线组于造模后第二天进行穴位埋线;模型组、穴位埋线组于造模后第9、13、18、23 d采集心脏血液并分离淋巴结中T淋巴细胞,进行流式细胞仪分析,同时对眼球进行组织病理切片。结果造模后第6至7 d大鼠眼部开始出现炎症反应,第13 d炎症达高峰(模型组临床评分为:3.42±0.58;穴位埋线组为:2.17±0.68,P=0.018,差异有统计学意义),之后逐渐开始消退。组织病理学结果显示模型组在第9、13、18、23 d时虹膜,睫状体及视网膜炎症均比穴位埋线组严重,而空白对照组眼部结构完整,未见明显炎症表现;流式细胞仪分析显示第13 d CD4+T占细胞比例最高值:模型组为85.97±1.03,穴位埋线组为:79.51±0.64,两组比较差异有统计学意义(P<0.001);第13 d时CD8+T最低:模型组为:14.03±1.03,组为:20.49±0.64,两组比较差异有统计学意义(P<0.001)。结论眼形态学、组织病理、细胞水平等结果显示,穴位埋线可有效减轻实验性自身免疫性葡萄膜炎大鼠炎症水平,对大鼠的葡萄膜炎具有治疗作用。OBJECTIVE To explore the therapeutic effect of catgut implantation at acupoint on rats with ex- perimental autoimmune uveitis （EAU）. METHODS Forty-eight liwes rat were randomly divided into control group, model group,＇ treatment group with 16 rats each. Control and treatment groups were immunized by IRBP, CFA,TB, the treatment group in second days after immunie by catgut implantation, All groups was collected blood and isolated lymphocyte after 9,13,18, 23 days, and then analyzed by flow cytometry. Further inflammation symptom was observed and confirmed by histopathologic examination in eye. RESULTS Uveitis inflammation was observed on day 6 or 7 and the severity peaked on dayl3 post immunization （Clinical mean score： EAU group 3.42±0.58, Catgut group 2.17± 0.68） and there was statistically significant differences （P=0.000）. After then the inflammation gradually subsided. Meanwhile, the morphologic change of organizational structure of iris and ciliary body also showed significant differ- ences between EAU group and catgut group on days 9, 13,18, 23. But there was no inflammatory cell with the iris and ciliary body on control group. The flow cytometry showed that CD4 T was the highest in T cells on day 13.EAU group： 85.97±1.03,catgut group： 79.51±0.64; CD8 T was the lowest in T cells on day 13. EAU group： 14.03± 1.03 ,catgut group： 20.49±0.64, and there was statisti cally significant differences （P=0.000）. COUCLU- SIONS The morphologic change of organizational structure on eye, the flow cytometry, the histopathology showed that there was a certain therapeutic effect of catgut implantation at acupoint on rats with experimental au- toimmune uveitis.
