摘 要:目的探讨甲钴胺对脑缺血再灌注大鼠脑组织细胞色素C表达的影响。方法选择Wistar大鼠132只,随机分为假手术组(24只)、模型组(27只)、尼莫地平组(27只)、甲钴胺低剂量组(27只)、甲钴胺高剂量组(27只)。采用改良线栓法栓塞大脑中动脉3h制作大鼠脑缺血再灌注模型;分别于6、12、24h用TUNEL法检测大脑皮质细胞凋亡、Western blot检测大脑皮质细胞色素C蛋白的表达。结果尼莫地平组、甲钴胺低、高剂量组6、12、24h阳性细胞数较模型组明显降低(P<0.01);模型组6、12、24h细胞色素C表达较假手术组明显升高(P<0.05,P<0.01);尼莫地平组、甲钴胺低、高剂量组6、12、24h细胞色素C表达较模型组明显降低(P<0.05,P<0.01);甲钴胺高剂量组6、12h细胞色素C表达较甲钴胺低剂量组明显降低(0.466±0.046 vs 0.617±0.033,P<0.01;0.163±0.081 vs 0.552±0.132,P<0.05)。结论甲钴胺对脑缺血再灌注损伤保护机制可能与抑制细胞色素C蛋白表达有关,以高剂量效果较好。Objective To study the effect of methycobal on expression of cytochrome C in brain tissue of rats following ischemia reperfusion(I/R)injury.Methods One hundred and thirty-two Wistar rats were randomly divided into sham operation group(n=24),model group(n=27),nimodipine treatment group(n=27),low methycobal dose treatment group(n=27),high methycobal dose treatment group(n=27).A rat I/R injury model was established by Longa occlusion of the middle cerebral artery.Apoptosis and expression of cytochrome C in brain tissue of rats were detected by TUNEL and Western blot respectively at 6,12,24 h.Results The number of positive cells was significantly smaller in nimodipine treatment group,low methycobal dose treatment group and high methycobal dose treatment group than in model group at 6,12,24h(P〈0.01).The expression level of cytochrome C was significantly higher in model group than in sham operation group,significantly lower in nimodipine treatment group,low methycobal dose treatment group and high methycobal dose treatment group than in model group at 6,12,24h(P〈0.05,P〈0.01)and in high methycobal dose treatment group than in low methycobal dose treatment group at 6,12h(0.466±0.046 vs 0.617±0.033,P〈0.01;0.163±0.081 vs0.552±0.132,P〈0.05).Conclusion Methycobal protects brain tissue of rats against I/R injury by downregulating the expression of cytochrome C.
